Title:Optimizacija ekstrakcije bioaktivnih komponent kurkume in formulacija v organogele : magistrsko delo

Abstract:Magistrsko delo prikazuje študijo optimizacij ekstrakcij bioaktivnih komponent iz kurkume in formuliranje ekstrakta v organogele. V laboratoriju smo izvajali ekstrakcijo po Soxhletu, hladno maceracijo, ultrazvočno ekstrakcijo in ekstarkcijo s superkritičnimi fluidi. Za pridobivanje ekstraktov sta bila uporabljena topila etanol in CO2, katera smo kasneje z rotacijskim uparjalnikom odparili. Ko smo pridobili vse željene ekstrakte je sledila analiza. Določili smo vsebnosti antioksidantov, totalnih fenolov in proantocianidinov z različnimi metodami pri različnih valovnih dolžinah. Izvedena je bila tudi tekočinska kromatografija z masno spekrometrijo LC-MS/MS. Iz dobljenih rezultatov smo ugotovili da imajo največjo vsebnost antioksidantov ekstrakti ultrazvočne ekstrakcije (64 % inhibicije) in ekstrakti pridobljeni s superkritičnimi fluidi (približno 1700 mg Trolox na g suhe mase). Ekstrakti superkritične ekstrakcije imajo prav tako največjo vsebnost totalnih fenolov (545 mg GA/100 g materiala), medtem ko imajo ekstrakti metode Soxhlet največjo vsebnost proantocianidinov (4,91 mg PAC/100 g materiala). Analiza LC-MS/MS je pokazala, da največ kurkumina vsebujejo ekstrakti, pridobljeni s postopkom ultrazvočne ekstrakcije (190,8 mg/g materiala), omenjen ekstrakt smo formulirali v organogele. Izdelana sta bila dva organogela; in sicer vodni ter brezvodni, nato je bila izvedena študija sproščanja kurkumina iz formulacij v simuliranem želodčnem soku. Ugotovljeno je bilo, da se pri nevodnem organogelu po času 240 min sprosti in raztopi večina kurkumina, medtem ko je delež sproščenega in raztopljenega kurkumina iz vodnega organogela v tem času veliko manjši in znaša zgolj okrog desetine (16 %).

Title:Optimization of extraction procedure for recovery of bioactive compounds from curcuma and its formulation into organogel
Abstract:The master thesis presents a study on optimization of extraction of bioactive components from turmeric and formulation of extracts in organogels. Soxhlet extraction, cold maceration, ultrasonic extraction and supercritical fluid extraction were performed. Ethanol was applied as a cosolvent in the case of szpercrital CO2 extraction. Remaining solvent residues were then evaporated using a rotary evaporator. After all the desired extracts were obtained, the analysis were performed: the content of antioxidants, total phenols, and proanthocyanidins was determined using UV/VIS spectrofotometry. Liquid chromatography with mass spectrometry - LC-MS /MS - was also performed. From the results, the highest content of antioxidants was found in the extracts obtained by ultrasonic extraction (64 % inhibition) and in the extracts obtained by supercritical fluid extraction (approximately 1700 mg Trolox per g dry weight). The extracts from supercritical fluid extraction also had the highest total phenolic content (545 mg GA/100 g material), while the extracts from the Soxhlet method had the highest content of proanthocyanidins (4,91 mg PAC/100 g of material). LC-MS/MS analysis showed that the extracts obtained by the ultrasonic extraction method had the highest curcumin content (190,8 mg/g material), and these extracts were formulated into organogels. Two organogels were prepared, one aqueous and one anhydrous, subsequent study of the release of curcumin from the formulations in simulated gastric acid was performed. It was found that in the non-aqueous organogel, most of the curcumin is released and dissolved after a time of 240 minutes, while the amount of curcumin released and dissolved from the aqueous organogel is much lower, with only about one-tenth (16%) of the curcumin released and dissolved in this time.
