Title:Kontrolirano sproščanje klorogenske kisline in njenih formulacij v simulacijskih raztopinah : magistrsko delo

Abstract:Klorogenske kisline so pomembne fenolne spojine, ki se jih pogosto najde v različnem sadju ter zelenjavi, daleč največjo vsebnost pa ima zelena kava. Te spojine imajo veliko pozitivnih učinkov, raziskave kažejo na protirakavo ter antioksidativno delovanje, prav tako pa pomagajo pri uravnavanju metabolizma lipidov ter sladkorjev. Sama ekstrakcija klorogenskih kislin iz njenih virov je otežena zaradi hitre degradacije ter izomerizacije klorogenskih kislin ter zmanjšane stabilnosti v raztopinah, zato se išče vedno nove ter izboljšane metode. Med uspešnejšimi metodami so bile metode LC-MS, ki smo jih za analizo vzorcev uporabili tudi mi. Za raziskovanje smo si izbrali 3-kofeoilkininsko kislino (3-CQA) , znano tudi pod imenom klorogenska kislina, saj je najpogostejša ter najbolj raziskana. Preverili smo njeno topnost v 0,1 M HCl, destilirani vodi, etanolu ter metanolu, ter ugotovili, da je bila največja topnost v etanolu, najmanjša pa v destilirani vodi. Nadalje smo preverili raztapljanje različnih oblik klorogenske kisline (v praškasti obliki oz. analitski standard, prehransko dopolnilo ter lastna formulacija stisnjene tabletke s klorogensko kislino in mikrocelulozo) tako v destilirani vodi kot v 0,1 M HCl. Najbolje se je izkazala kombinacija lastne formulacije (tabletke s klorogensko kislino in mikrocelulozo) v 0,1 M HCl, ki je pokazala tako visok delež raztopljenosti, do 90 % glede na zatehtano klorogensko kislino, kot tudi enakomerno rast deleža raztopljenosti po času raztapljanja. Raztapljanje analitskega standarda klorogenske kisline v destilirani vodi ni bilo uspešno, saj je delež raztopljenosti začel padati s časom raztapljanja, prav tako je bil enak problem pri raztapljanju prehranskega dopolnila v 0,1 M HCl. Raztapljanje praškaste oblike klorogenske kisline v 0,1 M HCl je bilo obetavno, s konstantnim višanjem raztopljenega deleža skozi ves čas raztapljanja. S klorogensko kislino v praškasti obliki smo izvedli stresne teste, in sicer v 1 M HCl, v 1 M NaOH, v 4 % H2O2, destilirani vodi ter v fosfatnem pufru s pH 7,4. Izkazalo se je, da je klorogenska kislina najstabilnejša v 1 M HCl ter zadovoljivo stabilna v 4 % H2O2, kar nakazuje na izredno dobro antioksidativnosti klorogenske kisline. V 1 M NaOH se je klorogenska kislina popolnoma razgradila že po 24 urah, v destilirani vodi ter fosfatnem pufru pa je deloma spremenila obliko ter prešla iz 3-CQA v 4-CQA ter 5-CQA, kar smo potrdili z LC-MS/MS metodo.

Title:Controlled release of chlorogenic acid and its formulations in simulation solutions
Abstract:Chlorogenic acids are important phenolic compounds, which are commonly found in different kinds of fruits and vegetables, green coffee having the biggest amount of them all. These compounds have a variety of positive effects, researches show anti-cancer and antioxidant properties, and are also shown to help regulate the metabolism of lipids and sugars. The extraction of chlorogenic acids from their sources itself is challenged because of the fast degradation and isomerization of chlorogenic acids, and because of the lowered stability in solutions, that is why newer and better methods are being researched. One of the more successful methods were LC-MS methods, which we also used for the analysis of our samples. For our researches we picked 3-coffeoylquinic acid (3-CQA), known under the commercial name chlorogenic acid because of it being the most common and most researched one. We checked the solubility of it in 0,1 M HCl, in distilled water, ethanol and methanol, finding it had the highest solubility in ethanol and the least soluble in distilled water. Furthermore, we checked the dissolution of different forms of chlorogenic acid (bought chemical in powder form, a dietary supplement and our own formulation of a pressed tablet containing chlorogenic acid and micro cellulose) in distilled water as well as in 0,1 M HCl, the best combination being our own formulation (tablets with chlorogenic acids) in 0,1 M HCl, which showed high percentage of dissolution, up to 90 % of the added chlorogenic acid, as well as a stable increase of the percentage of dissolution. The dissolution of the bought form of chlorogenic acid in distilled water wasn’t successful, as the percent of the dissolved compound kept decreasing as the dissolution went on time-wise, the same problem arising in the combination of the dietary supplement in 0,1 M HCl. The dissolution of the bought form of chlorogenic acid in 0,1 M HCl was promising, with steadily increasing percentage of dissolved compound throughout the whole dissolution process. Stress testing was done with the bought chlorogenic acid in powder form, namely in 1 M HCl, 1 M NaOH, 4 % H2O2, distilled water and in a phosphate buffer with pH 7,4. Results showed that chlorogenic acid was the most stable in 1 M HCl and had good stability in 4 % H2O2, which shows good antioxidant properties of chlorogenic acid. In 1 M NaOH, chlorogenic acid disintegrated wholly after 24 h, in distilled water and in the phosphate buffer, chlorogenic acid changed its form from 3-CQA to 4-CQA and 5-CQA.
