Abstract:
Namen magistrske naloge je bil določevanje antimikrobne učinkovitosti posameznih delov granatnega jabolka. Proučevali smo antimikrobno učinkovitost vzorcev iz različnih delov sadeža. Kot vzorce smo uporabili sveži sok, s Soxhletovo ekstrakcijo smo pripravili etanolna ekstrakta lupin in zrn, prav tako smo pripravili vodna ekstrakta lupin in zrn. Vzorce smo tudi liofilizirali in izvedli ekstrakcije iz liofiliziranih delov granatnega jabolka; vodna ekstrakta iz liofiliziranih lupin in liofiliziranih zrn ter etanolna ekstrakta liofiliziranih lupin in liofiliziranih zrn. Pridobljene ekstrakte iz svežih lupin in zrn ter iz liofiliziranih lupin in zrn smo liofilizirali z namenom koncentriranja dobljenih ekstraktov. Liofilizirali smo tudi sveži sok iz granatnega jabolka. Antimikrobno delovanje ekstraktov granatnega jabolka smo proučevali z disk difuzijsko metodo. Proučevali smo vpliv različnih ekstraktov, pridobljenih iz granatnega jabolka, na rast Escherishia coli, Pseudomonas fluorescens, Pseudomonas aeruginosa, Bacillus cereus, Aspergillus brasiliensis, Candida albicans. Najboljše inhibitorne lastnosti na rast E. coli izkazuje EtOH ekstrakt lupin. Pri koncentraciji E. coli 106 CFU/mL doseže premer inhibicije rasti mikroorganizma 18 mm. Največji premer inhibicije rasti P. fluorescens smo izmerili pri H2O ekstraktu zrn, ki je pri koncentraciji mikroorganizma 107 CFU/mL znašal 24 mm. Najboljše inhibitorne lastnosti na rast P. aeruginosa pri koncentraciji mikroorganizma 106 CFU/mL izkazuje H2O ekstrakt lupin, 16 mm. Na rast B. cereus izkazuje najboljšo antimikrobno aktivnost EtOH ekstrakt liofiliziranih lupin (23 mm) pri koncentraciji mikroorganizma 106 CFU/mL. Rast S. aureus najbolj zavira EtOH ekstrakt liofiliziranih lupin, saj pri koncentraciji mikroorganizma 105 CFU/mL znaša cona inhibicije rasti 19 mm. Prav pri nobenem testnem vzorcu granatnega jabolka ni bilo zaznati inhibitornega učinka na rast gliv. Visoke stopnje inhibicije bakterijskih kultur smo opazili pri EtOH ekstraktu lupin, H2O ekstraktu liofiliziranih lupin in liofiliziranemu H2O ekstrakt liofiliziranih lupin. Najslabše inhibitorne učinke na izbrane testne mikroorganizme izkazujeta H2O ekstrakt liofiliziranih zrn in liofilizirani H2O ekstrakti liofiliziranih zrn. Največ testiranih vzorcev granatnega jabolka je izkazovalo inhibitoren učinek na rast B. cereus, najmanj pa na rast P. aeruginosa.

Title:
Determination of antimicrobial efficiency of pomegranate
Abstract:
The aim of the master’s thesis was to determine the antimicrobial efficiency of individual parts of pomegranate. Samples from various parts of the fruit were prepared; fresh juice, aqueous peel, seed extracts, ethanol peel and ethanol seed extracts which were prepared by Soxhlet extraction were two lots of seed extract used. Samples were also lyophilized and extracted from lyophilized pomegranate parts: aqueous extracts from lyophilized peel and lyophilized seeds and ethanol seed extracts. The gained extracts from fresh peel and seeds and from lyophilized peel and seeds were lyophilized with the aim to concentrate the obtained extracts. We also lyophilized fresh pomegranate juice. The antimicrobial effect of pomegranates was determined by the disc diffusion method on various microbial cultures. The effect of the extracts obtained from pomegranate on the growth of Escherichia coli, Pseudomonas fluorescens, Pseudomonas aeruginosa, Bacillus cereus, Aspergillus brasiliensis, Candida albicans was studied. Ethanol speel extract shows the best inhibitory properties on E. coli growth, at a concentration of E. coli 106 CFU/mL it reaches a diameter of inhibition of microorganism growth of 18 mm. The maximum diameter of P. fluorescens growth inhibition was measured with aqueous seeds extract, which was 24 mm at a microorganism concentration of 107 CFU/mL. The best inhibitory properties on the growth of P. aeruginosa at a microorganism concentration of 106 CFU/mL show aqueous extract of peel, 16 mm. The best antimicrobial activity on the growth of B. cereus shows ethanol extract of lyophilized peel (23 mm) at a concentration of 106 CFU/mL. The widest diameter of S. aureus growth inhibition was measured with ethanol extract of lyophilized peel, which at a microorganism concentration of 105 CFU/mL is 19 mm. No test sample of pomegranate showed any inhibitory effect on yeast growth. High levels of inhibition on the growth of bacterial cultures using ethanol peel extract, aqueous extract of lyophilized peel and lyophilized aqueous extract of lyophilized peel were obserwed. The aqueous extract of lyophilized seeds and lyophilized aqueous extracts of lyophilized seeds show the lowest inhibitory effects of the selected test microorganisms. The different types of pomegranate samples had a greater inhibitory effect on the growth of B. cereus and least inhibitory effect on the growth of P. aeruginosa.
