Title:Uporaba encimskih skupkov za odstranjevanje diklofenaka iz odpadnih vod : diplomsko delo univerzitetnega študijskega programa I. stopnje

Abstract:Zamrežene encimske skupke (CLEAs) in zamrežene magnetne skupke (mCLEAs) smo pripravili iz encima lakaze. Imobilizacija je potekala ob prisotnosti mrežnega povezovalca – glutaraldehida (GA). Za pripravo mCLEAs smo uporabili maghemitne aminosilanske magnetne nanodelce. Pripravljenim CLEAs in mCLEAs smo po končanem postopku imobilizacije določili tudi učinkovitost imobilizacije na osnovi določene koncentracije prisotnih proteinov v supernatantu (s pomočjo Bradfordove metode). Namen diplomske naloge je bila presnova diklofenaka v njegove manj toksične produkte s pomočjo proste lakaze ter imobilizirane lakaze v obliki CLEAs in mCLEAs. Pri tem smo optimirali različne procesne parametre, kot so: koncentracija diklofenaka, temperatura in pH. Prav tako smo proučevali kinetiko reakcije ter stabilnost CLEAs in mCLEAs pri 4 °C. Za lakazo, imobilizirano v obliki CLEAs in mCLEAs, smo proučevali tudi možnost njene večkratne uporabe. Vsebnost dikofenaka v vzorcih smo analizirali z metodo tekočinske kromatografije visoke ločljivosti (HPLC). Določili smo optimalne pogoje za razgradnjo diklofenaka s prosto in imobilizirano lakazo v obliki CLEAs in mCLEAs. Na osnovi dobljenih rezultatov smo ugotovili, da encim lakaza v prosti obliki ter imobilizirani obliki CLEAs in mCLEAs uspešno razgrajuje diklofenak, vendar bi zaradi daljše stabilnosti, lažjega ločevanja in večkratne uporabe za razgradnjo diklofenaka na čistilne naprave aplicirali mCLEAs.

Title:Use of enzyme aggregates for removal of diclofenac from wastewater
Abstract:Cross-linked enzyme aggregates (CLEAs) and cross-linked magnetic aggregates (mCLEAs) were prepared from laccase enzyme. Immobilisation took place in the presence of cross-linking agent glutaraldehyde (GA). For preparation of mCLEAs we used maghemite aminosylane magnetic nanoparticles. After the process of immobilisation, the immobilisation efficiency of prepared CLEAs and mCLEAs was calculated based on determined protein concentrations in supernatant (with the help of Bradford's method). Purpose of diploma thesis was degradation of diclofenac to its less toxic products with help of free laccase enzyme and immobilised laccase in form of CLEAs and mCLEAs. Different processing parameters like: diclofenac concentration, temperature and pH were optimized. Reaction kinetics and stability of CLEAs and mCLEAs were also studied at 4 °C. For laccase, immobilised in CLEAs and mCLEAs, its ability of multiple use was studied. Content of diclofenac samples was determined with high-performance liquid chromatography (HPLC). Optimal conditions for diclofenac degradation with free and immobilised laccase in the form of CLEAs and mCLEAs were acquired. Based on results was determined, that laccase enzyme in free form and immobilised form of CLEAs and mCLEAs successfully decomposes diclofenac. mCLEAs would be more suitable to apply for diclofenac degradation on wastewater treatment plants, because of its longer stability, easier separation and reusability.
