Title:Vpliv sestave gojišča na aktivnost encimov iz Pleurotus ostreatus : diplomsko delo univerzitetnega študijskega programa I. stopnje

Abstract:V diplomskem delu je opisano gojenje glive Pleurotus ostreatus na različnih gojiščih, ki so bila sestavljena iz mešanice pšeničnih otrobov ter sezonskega sadja in zelenjave. Proučevali smo vpliv sestave gojišča na prirast glive ter na aktivnost encimov (lakaze, celulaze in α-amilaze) v ekstraktih po 8 dneh gojenja glive. Razrast glive smo spremljali 8 dni in v določenih intervalih spremljali prirast oz. zmanjšanje biomase. Po 8 dneh smo ekstrahirali encime iz razrastlega P. ostreatus na izbranih gojiščih. Določili smo koncentracijo proteinov ter aktivnost in specifično aktivnost posameznih encimov (lakaze, celulaze in α-amilaze) v encimskih ekstraktih. Nato smo iz encimskih ekstraktov pripravili zamrežene encimske skupke (CLEAs). Določili smo učinkovitost imobilizacije, nato pa še aktivnost in specifično aktivnost za vse 3 encime v CLEAs. Zanimala nas je tudi stabilnost CLEAs, ki smo jo skladiščili pri 4°C. Preostalo aktivnost CLEAs smo zasledovali po 2 in 4 tednih skladiščenja pri 4°C.

Title:The influence of composition of the medium of enyzmes activity Pleurotus ostreatus
Abstract:The diploma work describe the cultovation of Pleurotus ostreatus on various residues of fruits and vegetables, in addition to wheat bran. The fungus growth was observed for 8 days and weight in between to measure the weight of the fungus. After 8 days, we did the enzyme extraction. The enzymes like cellulase, laccase and α-amylase were observed and the protein concentration was determined according to the Bradford method. Activity and specific activity in enzyme extract were also determined. Subsequently, cross-linked enzyme aggregates (CLEAs) were made. The immobilization efficiency was determined, and then the residual activity and residual specific activity for all 3 enzymes were measured. We were also interested in the stability of enzymes in CLEAs over time at 4°C, so we set cross-linked enzyme aggregates at 4°C and measured the enzyme activity after 2 and 4 weeks.
