Abstract:
V diplomski nalogi je prikazana študija aktivnost posameznih encimov v pralnih sredstvih. Za študijo smo izbrali praškaste pralne praške, tekoče pralne praške in koncentrirane pralne praške, ki so na voljo v obliki pralnih kapsul. Osredotočili smo se predvsem na pralna sredstva, ki so na voljo na slovenskem tržišču. Pri delu smo uporabili tako praške, pri katerih deklaracija navaja vsebnost encimov, kot na take, kjer deklaracija vsebnosti encimov ne navaja. Pri delu smo poskušali posnemati princip pranja v pralnem stroju, zato smo za potrebe študije, vnaprej pripravljeno raztopino trdnega ali tekočega pralnega sredstva 1 uro inkubirali pri različnih temperaturah 30 °C, 40 °C, 60 °C in 95 °C. V tako pripravljenih raztopinah pralnih sredstev smo s pomočjo UV - Vis spektrofotometra določili koncentracijo skupnih proteinov in aktivnosti encimov α-amilaze, celulaze, lipaze in proteaze. Ugotovili smo, da so našteti encimi najpogosteje zastopani v pralnih sredstvih. Rezultati kažejo, da je v večini pralnih sredstev aktivna proteaza. Najvišje aktivnosti v pralnih sredstvih dosega encim lipaza, medtem ko je najmanj aktiven encim celulaza, katerega višje aktivnosti smo določili samo v štirih primerih. Določene aktivnosti encimov v pralnih sredstvih so relativno nizke. Posamezni encimi, ki so dodani v pralnih sredstvih imajo različne temperaturne optimume. Glede na to lahko sklepamo, da je posamezen prašek različno učinkovit pri različnih temperaturah ob enakem časovnem obdobju pranja.

Title:
Enzyme activity in liquid and solid detergents
Abstract:
The thesis shows the study of the contents of enzymes and their activities in detergents. In this study, we selected a powder laundry detergents, liquid laundry powders and concentrated washing powders, which are available in the form of washing capsules. We focused primarily on detergents, which are available on the Slovenian market, namely powders containing enzymes and such that enzyme is not contained. In part, we try to emulate the principle of washing in the washing machine, so we need studies, pre-prepared solution of solid or liquid detergent incubated 1 hour at different temperatures - at 30 °C, 40 °C, 60 °C and 95 °C. In such prepared solutions of washing detergents, the concentration of total proteins and the activity of α-amylase enzymes, cellulase, lipase and protease enzymes was determined using UV-Vis spectrophotometer. We found that the enzymes listed are most often represented in detergents. The results show that in most detergents the active protease is active. The highest activity in detergents reaches the lipase enzyme, while the least active cellulase enzyme, whose higher activity was determined only in four cases. Certain activities of enzymes in detergents are relatively low. Individual enzymes added to detergents have different temperature optimms. According to this, it can be concluded that the individual powder is at different temperatures differently effective with the same washing time period.
