Abstract:
Probiotiki so živi mikroorganizmi, ki ob zaužitju v zadostnih količinah ugodno vplivajo na zdravje gostitelja. Mehanistično gledano izboljšajo ravnovesje črevesne mikrobiote in funkcijo črevesja, okrepijo imunski odziv, branijo črevesje pred patogenimi bakterijami, imeli naj bi tudi protitumorske učinke. V naši raziskavi smo skušali določiti protitumorski vpliv izbranih probiotičnih mikroorganizmov in njihovih metabolitov na viabilnost tumorskih črevesnih epitelnih celičnih linij AGS in CaCo-2, za kontrolo pa smo imeli normalne črevesne epitelne celice linije HIEC. V ta namen smo uporabili probiotične kulture Lactobacillus plantarum PCS20 in PCS22, Lactobacillus rhamnosus GG (LGG), Lactobacillus paracasei (Shirota) in Lactobacterium spp PCK9. Za negativno kontrolo smo uporabili nepatogeno Escherichio coli (E. coli). Bakterije in njihove supernatante smo nanašali v različnih koncentracijah na celice v vodnjake na ploščici P-96 in inkubirali 24 ur. Po pretečenem inkubacijskem času smo rezultate viabilnosti celic pridobili z označevanjem celic s kristal vijoličnim barvilom. Rezultate viabilnosti celic smo vnesli v Excel, jih ovrednotili in grafično prikazali. Protitumorsko delovanje probiotikov smo uspeli dokazati le delno, saj smo ugotovili, da je vpliv bakterij in njihovih metabolitov odvisen od vrste probiotičnih bakterij in njihovih supernatantov, njihovih koncentracij in od uporabljene humane celične linije.

Title:
CITOTOXICITY OF PROBIOTICS AND THEIR METABOLITES ON GUT EPITHELIAL CELLS
Abstract:
Probiotics are live microorganisms, which upon administration in adequate amounts confer health benefits to the host. From the mechanistic point of view they improve the balance of intestinal microbiota and bowel function, enhance and modulate immune response, defend the gut against pathogenic bacteria and they are also claimed to have anti-tumor effects. In our study we sought to determine the anti-tumor effect of selected probiotic microorganisms and their metabolites on the viability of intestinal epithelial tumor-derived cell lines AGS and CaCo-2. As a control, normal intestinal epithelial cell line HIEC was used. As of probiotics, bacterial strains Lactobacillus plantarum PCS20 and PCS22, Lactobacillus rhamnosus GG (LGG), Lactobacillus paracasei (Shirota) and Lactobacterium spp. PCK9 were tested. Non-pathogenic Escherichia coli (E. coli) served as a negative control. Bacteria and their supernatants were applied in different concentrations on cells grown in the wells of P-96 plate for 24 h. After the incubation, the results of cell viability were obtained by crystal violet staining of alive cells. The results were evaluated and presented by Excel charts. Anti-tumor activity of some probiotic bacteria and their supernatants was proved only partially, since cytotoxic effects depended both on the bacterial strain and their supernatants, applied concentrations as well as the human cell line used in the experiment.
