Abstract:
Diplomsko delo je sestavljeno iz dveh delov. Najprej smo sintetizirali magnetne zamrežene encimske skupke iz celulaze. Pri tem smo optimizirali parametre, kot so hitrost stresanja na stresalniku, koncentracijo mrežnega povezovalca glutaraldehida, čas stresanja in temperaturo, z namenom doseči čim večjo učinkovitost imobilizacije in preostalo aktivnost imobilizirane celulaze. Postopek priprave magnetnih zamreženih skupkov sestoji iz dveh korakov, obarjanja in zamreženja. Kot obarjalni reagent smo uporabili metanol. V drugem delu diplomske naloge, smo magnetne zamrežene encimske skupke (mCLEAs) iz celulaze sintetizirali pri optimalnih pogojih in jih skladiščili pri 23°C. Zanimalo nas je, kako izbrana temperatura in čas skladiščenja vplivata na preostalo aktivnost mCLEAs. Proučevali smo tudi vpliv tlaka pri konstantni temperaturi na preostalo aktivnost mCLEAs. mCLEAs smo izpostavili superkritičnemu ogljikovemu dioksidu pri 100 bar in 200 bar pri različnih časih izpostavitve.

MAGNETIC CROSS LINKED ENZYME AGGREGATES FROM CELLULASE
Abstract:
This thesis is composed of two parts. Firstly, we synthesized magnetic cellulase cross-linked enzyme aggregates. We optimized parameters like agitation rate on the shaker, concentration of glutaraldehyde, agitation time and temperature to achieve better immobilization efficiency and activity of immobilized cellulase. The preparation of magnetic cross-linked aggregates consists of two steps, precipitation and crosslinking. We used methanol as precipitation reagent. In the second part of the thesis we synthesized magnetic cellulase cross-linked aggregates (mCLEAs) at optimal conditions and stored them at 23°C. We wanted to know how a specific temperature and storage time would affect the mCLEAs activity. We also studied the effect of pressure at constant temperature on mCLEAs activity and exposed the mCLEAs to supercritical carbon dioxide at 100 and 200 bars at different times of exposure.
