Abstract:
Emulgator je snov, ki povzroča in pospešuje nastanek emulzije. Emulgatorji sodijo med površinsko aktivne snovi. Imajo značilno kemijsko zgradbo, del molekule je hidrofilen (topi se v vodi), drug del pa hidrofoben (topi se v maščobah). Na tak način so emulgatorji razporejeni med vodno in oljno fazo in znižujejo medfazno napetost, ki je glavni vzrok nestabilnosti emulzij. Polisorbati so skupina emulgatorjev, ki se uporabljajo v živilski, farmacevtski in kozmetični industriji, po videzu so mastne tekočine, narejene iz derivata sorbitola in maščobnih kislin. Na trgu se pojavljajo s tržnimi imeni Alkest, Canarcel in Tween. V sestavi na etiketi živil so označeni z oznako E431-E436, polisorbat 80 ima E številko 433. Za namene razvoja in raziskav v podjetju Vitiva smo optimirali in validirali HPLC metodo, ki omogoča ugotavljanje vsebnosti polisorbata 80 v pripravkih, ki vsebujejo izvleček rožmarina in v naravnih barvilih. Kromatografske pogoje smo optimirali in validirali, določili smo specifičnost, linearnost, natančnost, meji zaznave in določanja, točnost in robustnost metode. Dokazali smo, da je predlagana analizna HPLC metoda za ugotavljanje vsebnosti polisorbata primerna, natančna in točna. Ugotovili smo, da je standardna raztopina polisorbata stabilna, razpada le pri izpostavljenosti sončni svetlobi.

DETERMINATION OF EMULGATOR POLYSORBATE BY HIGH PERFORMANCE LIQUID CHROMATOGRAPHY
Abstract:
Emulsifiers are substances that cause and promote the formation of emulsions. They are surfactant substances with typical chemical structure; part of the molecule is hydrophilic (water soluble) and the other part is hydrophobic (fat soluble). Emulsifiers are dispersed into water and oil phases and lower the interphase tension - main reason for emulsion instability. Polysorbates are a group of emulsifiers used in the food, pharmaceutical and cosmetics industries. Polisorbates are oily liquids derived from a derivative of sorbitol and fatty acids. Common brand names they appear under are Alkest, Canarcel and Tween. On food labels they are listed under E431-E436. Polysorbate 80 is listed under E433. For research and development purposes in the company Vitiva the HPLC method was optimized and validated, which enables the determination of Polysorbate 80 in compounds containing rosemary extract and natural colorauts. The chromotopgraphic conditions were optimized and validated. Specificity, linearity, accuracy, detection limit, precision and robustness of the method was determined. It was proved that the proposed HPLC analysis method for the determination of polysorbate 80 is appropriate, precise and accurate. The standard solution of polysorbate is stable; it degrades only when exposed to sunlight.
