Title:PLINSKA KROMATOGRAFIJA Z MASNO SPEKTROMETRIJO – PRIMER KVALITATIVNE ANALIZE PROSTIH MAŠČOBNIH KISLIN V KRVNI PLAZMI ČLOVEKA

Abstract:V diplomskem delu obravnavamo plinsko kromatografijo v povezavi z masno spektrometrijo in njeno aplikacijo na kvalitativno določanje vsebnosti prostih maščobnih kislin v krvni plazmi človeka. Diploma je sestavljena iz dveh delov. V teoretičnem delu na splošno predstavimo masno spektrometrijo in kromatografijo, pri čemer dajemo poudarek masni spektrometriji v povezavi s plinsko kromatografijo, saj so na takšni napravi opravljene kasnejše meritve. Opišemo najpogosteje uporabljene metode priprave vzorca, injiciranja, kromatografije, ionizacije in masne analize. V praktičnem delu pa opišemo postopke analize prostih maščobnih kislin v krvni plazmi in meritve s plinskim kromatografom in masnim spektrometrom v lasti Fakultete za naravoslovje in matematiko. Oprema je bila kupljena v okviru Norveškega finančnega mehanizma. Z meritvami določimo masne spektre estrov maščobnih kislin v referenčnem vzorcu, ki vsebuje 37 različnih komponent. Identificiramo jih 23 od 37. Nato analiziramo vzorce, pridobljene iz krvne plazme človeka in opravimo primerjavo. Vzorce pripravimo po dveh načinih, pri čemer ločimo že v plazmi esterificirane in neesterificirane proste maščobne kisline. Na podlagi primerjave izmerjenih masnih spektrov z referenčnimi masnimi spektri iz knjižnice in iz referenčne meritve pokažemo, katere proste maščobne kisline so vsebovane v vzorcu krvne plazme. Z meritvami pokažemo, da v obeh vzorcih neznanih darovalcev krvi najdemo le tri proste maščobne kisline in da so le-te v obeh primerih enake. Hkrati tudi opazimo, da v krvni plazmi ni prisotnih esterificiranih prostih maščobnih kislin oz. potrdimo, da je njihova morebitna vsebnost pod mejo detekcije naprave.

Title:GAS CHROMATOGRAPHY WITH MASS SPECTROMETRY – AN EXAMPLE OF QUALITATIVE ANALYSIS OF FREE FATTY ACIDS IN HUMAN BLOOD PLASMA
Abstract:The thesis deals with gas chromatography in relation to mass spectrometry and its application in the qualitative analysis of the degree of free fatty acids in human blood plasma. The thesis is composed of two parts. The theoretical part gives a general overview of mass spectrometry and chromatography with an emphasis on mass spectrometry in relation to gas chromatography. This methodology is used also in the practical part of the thesis. The most commonly used methods for sample preparation, injection, chromatography, ionization and mass spectrometry are also presented in the theoretical part. The practical part of the thesis focuses on the procedures for analysing free fatty acids in human blood plasma and measurements performed with a mass spectrometer and gas chromatograph, which belongs to the Faculty of Natural Sciences. The acquisition of the equipment for the thesis was financed by the Norway Grants. Based on measurements, the mass spectra of esterified fatty acids are determined in the reference sample containing 37 different components. 23 of 37 free fatty acids are identified. The samples obtained from human blood plasma are then analysed and compared to the reference sample. Samples are prepared following two different methods by separating eseterified free fatty acids and non-esterified free fatty acids in human blood plasma. The comparison between the measured mass spectra and the reference mass spectra in the database and further with reference measurements indicates the presence of free fatty acids in the sample of human blood plasma. The analysis of two different samples of human blood plasma reveals the presence of only three different free fatty acids. At the same time the analysis also reveals that esterified free fatty acids are not present in the blood plasma, or at least not to a degree that they could be detected.
