Abstract:
Med je hranljivo živilo, ki ga uživajo po vsem svetu in spremlja človeško prehrano že iz davnih časov. Njegovo konzumiranje je povezano predvsem z zdravim načinom življenja, zaradi pozitivnih lastnosti, ki spodbujajo zdravje. Poleg koristnih bioaktivnih spojin v medu, kot so fenoli, pa je mogoče najti tudi nekatere s potencialno škodljivimi učinki, kot so pirolizidinski alkaloidi (PA). PA sestavljajo raznoliko skupino, približno 660 struktur, sekundarnih metabolitov, ki predstavljajo pomembno vlogo pri konstitutivni kemični obrambi rastlin pred rastlinojedci. Veljajo za ene izmed najbolj razširjenih naravnih toksinov, ki ponazarjajo tveganje za zdravje ljudi, saj so zastrupitve povezane z akutnimi in kroničnimi poškodbami jeter in lahko povzročijo smrt. V zadnjih letih se tako pojavlja skrb vzbujajoče vprašanje glede varnosti živil, saj uživanje izdelkov, ki vsebujejo PA, predstavlja potencialno tveganje za zdravje potrošnikov. Zdi se, da je stopnja onesnaženosti medu s PA odvisna od geografskega in botaničnega izvora. Zato smo se v magistrskem delu osredotočili na določitev in identifikacijo pirolizidinskih alkaloidov in njihovih N-oksidov v različnih vrstah slovenskega medu (cvetlični, ajdov, gozdni, akacijev, lipov in kostanjev med). Kot dopolnilo k ocenjevanju vzorcev pa je bila izvedena tudi identifikacija v cvetličnem medu različnega geografskega porekla. Vsebnost PA v medu smo določili s ciljno analizo z uporabo sistema LC-MS/MS s predhodnim čiščenjem vzorcev s SPE.

Title:
Determination of pyrrolizidine alkaloids in honey
Abstract:
Honey is a nutritious food that is consumed all over the world and has been part of the human diet since ancient times. Its consumption is mainly linked to a healthy lifestyle, thanks to its positive health properties. In addition to the beneficial bioactive compounds in honey, like phenols, it is also possible to find some molecules with potentially harmful effects, such as pyrrolizidine alkaloids (PAs). PAs constitute a diverse group of approximately 660 structures, secondary metabolites, which play an important role in the chemical defense of plants against herbivores. They are considered to be among the most widespread natural toxins, illustrating the risk to human health. Poisoning is associated with acute and chronic liver damage and can cause death. Thus, in recent years, there has been an emerging concern about food safety, as the consumption of products containing PAs poses a potential health risk to consumers. The level of PA contamination in honey appears to depend on the geographical and botanical origin. Therefore, in our study we focused on the determination and identification of pyrrolizidine alkaloids and their N-oxides in different Slovenian honeys (floral, buckwheat, forest, acacia, lime and chestnut honeys). As a complement to the evaluation of the samples, identification was also carried out in floral honeys of different geographical origins. The PA content of the honeys was determined by targeted analysis using LC-MS/MS with pre-purification of the samples by SPE.
