Title:Vpliv prehrane in starosti goveda na kolonizacije s Clostridioides difficile : magistrsko delo

Abstract:Clostridioides difficile je anaerobna, sporogena bakterija, ki ob porušeni normalni črevesni flori proizvaja toksine in tako povzroči širok spekter črevesnih okužb pri ljudeh in živalih. V okviru magistrskega dela smo želeli raziskati vpliv spremenjene celotne krmne mešanice za govedo (TMR) na kolonizacije s C. difficile v fekalni mikrobioti 30-ih krav molznic glede na zaporedno laktacijo (1.‒6.) in fazo laktacije (I., II., III.). Prav tako smo v raziskavo vključili tudi 32 telet iz 6-ih starostnih skupin, da bi proučili, ali obstaja vpliv starosti in prehrane na kolonizacije s C. difficile v telečjem fecesu. Skupno smo 92 vzorcev inkubirali v bogatitvenem gojišču BHI z dodanim L-cisteinom, kvasnim ekstraktom in tauroholatom (BHIST) in kulturo po alkoholnem šoku inkubirali na selektivnem kromogenem gojišču. Analizo smo izvedli v laboratoriju Katedre za mikrobiologijo na Medicinski fakulteti Univerze v Mariboru. Noben fekalni vzorec ni bil pozitiven na bakterijo C. difficile. Na podlagi rezultatov sklepamo, da na kmetiji, zajeti v raziskavo, bakterija C. difficile v fecesu krav molznic in telet ni prisotna.

Title:Influence of Diet and Cattle Age on Colonizations with Clostridioides difficile
Abstract:Clostridioides difficile is an anaerobic, spore-forming bacterium that produces toxins when the normal intestinal flora is disrupted, causing a wide range of intestinal infections in humans and animals. This work aimed to investigate the influence of altered total mixed rations (TMR) on C. difficile colonies in the faecal microbiota of 30 dairy cows, according to successive lactations (1st‒6th) and lactation phases (I., II., III.). We also included 32 calves from 6 age groups in the study to determine if there was an effect of age and diet on colonizations with C. difficile in calf faeces. A total of 92 samples were incubated in an enrichment BHI broth with L-cysteine, yeast extract and taurocholate added (BHIST), subjected to alcohol shock and isolated on chromogenic plates. The analysis was performed in the laboratory of the Department of Microbiology at the Medical Faculty of the University of Maribor. Overall, no sample was positive for C. difficile. Our results indicate that C. difficile is not present in the faeces of dairy cows and calves on the farm included in the study.
