Title:Izolacija biološko aktivnih komponent iz plodov gozdnih borovnic ( Vaccinium myrtillus L. )

Abstract:Gozdne borovnice zaradi mnogih zdravilnih lastnosti, ki jih imajo na telo, sodijo med tako imenovana »superživila«. V magistrskem delu smo raziskali, kakšna je biološka učinkovitost ekstraktov plodov gozdnih borovnic. Osredotočili smo se predvsem na izolacijo fenolnih spojin. Zanje velja, da v kombinaciji z drugimi biološko aktivnimi spojinami, prisotnimi v rastlinskih ekstraktih, delujejo antioksidativno, protivnetno, protimikrobno in prav tako zdravijo oziroma preprečujejo nastanek in razvoj rakavih celic. Ekstraktom smo spektrofotometrično določili vsebnost skupnih fenolov, proantocianidinov in antioksidativno učinkovitost. S kromatografsko analizo (LC-MS/MS) smo kvalitativno in kvantitativno določili vsebnost posameznih fenolnih spojin v ekstraktih. Ker so rastlinski ekstrakti v vodi slabo topni, smo njihovo biološko dostopnost povečali z vezavo na biorazgradljiv polimerni nosilec s PGSS™ metodo. Morfološke lastnosti (velikost delcev in poroznost) smo določili z vrstičnim elektronskim mikroskopom (SEM). V magistrskem delu smo iz plodov borovnic uspešno izolirali fenolne spojine in ugotovili, da na kakovost ekstrakta vpliva izbira ekstrakcijske metode in vrsta topila. Ker so fenolne spojine polarne, je za njihovo izolacijo primerneje uporabiti polarna topila, kot so voda, metanol in etanol. Ugotovili smo, da je antioksidativna učinkovitost ekstraktov pogojena z vsebnostjo fenolnih spojin. V ekstraktih borovnic je bil najvišji delež klorogenske kisline. Klorogenska kislina deluje protibakterijsko napram S. aurerus in E. coli, kar smo v magistrskem delu uspešno dokazali. Prav tako je bila uspešno izvedena formulacija ekstraktov na polimernim nosilec.

Title:Isolation of biologically active compounds from bilberries (Vaccinium myrtillus L.)
Abstract:Bilberries, belong to the so-called group of “superfoods” due to the beneficial properties they have on the health. In the present master thesis, the biological efficiency of bilberry fruit extracts has been examined. Isolation of phenolic compounds has been carried out due to their antioxidant, anti-inflammatory and antimicrobial action. Phenolics, in combination with other biologically active compounds, act as inhibitors for the development of cancerous cells. The content of total phenolics, proanthocyanidines and the antioxidative activity has been determined spectrophotometrically. Quantitative and qualitative determinations of individual phenolic compounds in the extracts were performed by chromatographic analysis (LC-MS/MS). Most plant extracts are poorly soluble in water and are therefore known of their low biological availability. In order to increase their bioavailability, the extracts have been loaded on a polymer carrier with the PGSS™ method. The morphological properties (particle size and porosity) were determined by scanning electron microscopy (SEM). In this work, the isolated phenolic compounds from bilberry fruits were successfully isolated. It was observed that the quality of the extract is mostly dependent on the extraction method and solvent type. Polar solvents, such as water, methanol and ethanol were preferred solvents for the isolation of phenolic compounds, because of their polarity. It was discovered, that the phenolic compound content largely influenced the antioxidative activity. Chlorogenic acid was the major among the detached phenolic compounds in the bilberry extracts. This work demonstrated antibacterial effect of extracts containing chlorogenic acid against S. aurerus and E. coli. We have also successfully micronized our extract with a polymer carrier.
