Title:SOČASNO DOLOČANJE IZBRANIH FLAVONOIDOV V RASTLINSKIH EKSTRAKTIH S HPLC

Abstract:Validirali smo analizno metodo za sočasno določanje izbranih flavonoidov s tekočinsko kromatografijo visoke ločljivosti z ultravijolično zaznavo (HPLC-UV). Sočasno smo določali katehin, rutin, kvercetin in krisin. Spojine smo eluirali gradientno, pri čemer smo za mobilni fazi uporabili metanol in vodo nakisano z ocetno kislino. Separacijo in kvantifikacijo smo dosegli v 21 minutah pri valovni dolžini 280 nm. Z izbranimi validacijskimi parametri smo preverjali ustreznost metode. Rezultate smo statistično obdelali in določili ponovljivost, natančnost, linearnost, mejo zaznavnosti (LOD) in mejo določljivosti (LOQ). V diplomskem delu smo želeli razviti optimalno metodo za določanje izbranih flavonoidov v različnih vzorcih prehranskih dopolnil. Vzorce smo pripravili z ekstrakcijo trdno-tekoče. Raztopili smo jih v metanolu, ekstrahirali na ultrazvočni kopeli in filtrirali skozi filter papir in filtrno membrano ter analizirali. S to metodo nismo zaznali izbranih flavonoidov v prehranskih dopolnilih, zato smo po ekstrakciji na ultrazvočni kopeli zatehte vzorcev povečali in jih ekstrahirali s pomočjo rotacijskega uparjalnika pri 45 ºC, 1 uro. Tudi s to metodo nismo določili izbranih flavonoidov. Spojine smo želeli izolirati tudi s pomočjo ekstrakcije na trdni fazi (SPE). V ta namen smo preizkusili več različnih sorbentov. Kot najučinkovitejši se je pokazal hidrofilno-lipofilni kopolimer (HLB).

Title:SIMULTANEOUS DETERMINATION OF SELECTED FLAVONOIDS IN PLANT EXTRACTS WITH HPLC
Abstract:Analytical method for the simultaneous determination of selected flavonoids with high-performance liquid chromatography with ultraviolet detection (HPLC-UV) was validated. Catechin, rutin, quercetin, and chrysin were determined. The compounds were separated by gradient elution, whereas in the mobile phase, methanol and water acidified with acetic acid was used. The separation and quantification were achieved in 21 minutes at a wavelength of 280 nm. With the selected validation parameters, was confirmed the suitability of the method. The results were statistically processed in order to determine the repeatability, accuracy, linearity, limit of detection (LOD), and limit of quantitation (LOQ). The main goal of this work was developing of an optimal method for the determination of selected flavonoids in various plant samples and food supplements. The samples were prepared with the solid-liquid extraction. They were dissolved in methanol and extracted in an ultrasonic bath, and before analysis filtered through paper and a filter membrane. This method did not detect the selected flavonoids in the food supplements. Because that reason after the ultrasonic extraction the weight of the samples were increased and extraced on a rotary evaporator at 45 ºC for 1 h. Also this method did not determine the selected flavonoids. For isolation of investigated compounds also solid phase extraction (SPE) using different sorbets was tested. Hydrophilic-lipophilic copolymer has been proven to be most effective.
