Abstract:
V sklopu magistrskega dela smo preučevali razvoj in validacijo analizne metode za določevanje oksalatnih ionov v različnih rastlinskih vzorcih z uporabo ionske kromatografije. Sočasno z oksalatnimi ioni smo določili v istih vzorcih še vsebnosti kloridnih, nitratnih in sulfatnih ionov. Razvito metodo smo tudi delno validirali, z namenom potrditve ustreznosti izbrane metode. Določili smo linearnost, mejo določljivosti, mejo zaznavnosti, natančnost in pravilnost. V obseg validacije smo vključili tudi stabilnostno študijo standardov. Rezultati kažejo na ustreznost razvite metode, ki daje ponovljive, obnovljive, natančne in točne rezultate v ustreznem delovnem območju izbranih anionov. Topne oksalate smo iz vzorcev ekstrahirali z vodo na vodni kopeli pri 80 °C in 15 min. V raziskavo smo vključili rabarbaro (olupljeno steblo in olup), krompir, gomolj zelene, pastinak, korenje, jagode, liste bezga in korme tara. Med analiziranimi vzorci je bila vsebnost vodotopnih oksalatov najvišja v rabarbari. V olupu smo določili nekoliko višjo vsebnost (473 mg/100 g sveže mase) kot v steblu (331 mg/100g sveže mase). Vrednosti v preostalih vzorcih so bile nižje od 1,2 mg/100 g sveže mase za vzorec jagod do 15,9 mg/100 g sveže mase za vzorec korenja. V bezovih listih in kormih tara so bile vsebnosti vodotopnih oksalatov 89,5 mg/100 g suhe snovi (vzorci bezgovih listov) in med 104 ter 134 mg/100 g suhe snovi (kormi tara).

Title:
THE DETERMINATION OF OXALATE IONS IN PLANT SAMPLES USING ION CHROMATOGRAPHY
Abstract:
The master's thesis explores the development and validation of the analytical method for determining the oxalate ion content in different plant samples using ion chromatography. In addition to oxalate ions, the content of chloride, nitrite and sulphate ions was determined in the same samples. Special emphasis was placed on the selection and development of an adequate method for identifying and quantifying the ions. The effect of the different mobile phase compositions, the effect of the flow rate on the ion retention times, and the consequent changes in the column pressure were investigated. Thus, the research aimed to achieve the adequate peak separation in the shortest analysis time possible. The developed method was partially validated in order to confirm its adequateness. Linearity, limit of quantification, limit of detection, precision and trueness have been established. The stability study of standards was also included in the validation process. The results suggest the developed method is adequate since it renders repeatable, reproducible, precise and accurate results in the appropriate range of the selected anions. Water soluble oxalates were extracted on water bath at 80 °C for 15 min. Samples included in this research were: rhubarb (skin and peeled stalk), potatoes, celery root, parsnip, carrots, strawberries, elder tree leaves, and taro corms. Among the analysed samples, the highest oxalate concentrations were found in rhubarb. The oxalate content in the skin was slightly higher (473 mg/100 g FW (fresh weight)) than in the peeled stalks (331 mg/100 g FW). The oxalate contents determined in the rest of samples were in the range from 1.2 mg/100 g FW in strawberries to 15.9 mg/100 g FW in the carrot. The elder’s leaves and taro corms contained relatively low levels of soluble oxalates 89.5 mg/100 DW (dry weight) for elder’s leaves and from 104 mg/100 g DW to 134 mg/100 g DW for taro corms.
