Title:Optimizacija encimatske hidrolize tekstilnih odpadkov iz mešanice poliestra in celuloze

Abstract:Tekstilni odpadki oz. odpadni material predstavlja velik in rastoč problem v današnji potrošniški družbi, zato smo se v tej diplomski nalogi osredotočili na encimatsko hidrolizo celuloze v materialu, ki vsebuje dve tretjini polietilen tereftalata in eno tretjino celuloze. Preizkusili smo različne pogoje, torej kakšen je vpliv različnih temperatur, koncentracij encimske mešanice (Cellic® CTec2) in dveh različnih pufrskih raztopin. V prvem delu poskusa smo testirali, ali 40 % povečanje količine encimske mešanice pomeni hitrejšo razgradnjo celuloze oziroma višjo koncentracijo glukoze v vzorcih. Ugotovili smo, da večja količina encimske mešanice pomeni malo boljše rezultate, ampak je ta razlika minimalna in nezadostna, da bi bila uporaba večje količine ecimske mešanice smotrna. Prav tako smo preverili, kako reakcija poteka v prisotnosti 0,1 M pufrske raztopine (NH4)2SO4 in 0,1 M pufrske raztopine NH4Cl. Rezultati so pokazali, da je precej več glukoze nastalo v pufrski raztopini (NH4)2SO4 in tudi poraba NaOH je bila veliko manjša. V drugem delu poskusa pa nas je zanimalo, kakšen vpliv na reakcijo in encimsko mešanico imajo tri različne temperature, in sicer 50, 55 in 60 °C. Iz podatkov je razvidno, da je najoptimalnejša temperatura 50 °C, saj je v tem reaktorju nastalo največ glukoze.

Title:Optimization of encimatic hydrolysis of textile waste composed of polyester and cellulose
Abstract:Textile waste represents a significant and growing problem in today's consumer society. Therefore, this thesis focuses on the enzymatic hydrolysis of cellulose in a material composed of two-thirds polyethylene terephthalate and one-third cellulose. We tested various conditions to determine the effects of different temperatures, concentrations of the enzyme mixture (Cellic® CTec2), and two different buffer solutions. In the first part of the experiment, we investigated whether a 40% increase in the enzyme mixture would result in faster cellulose degradation and in a higher concentration of glucose in the samples. We found that a higher amount of enzymes yielded only slightly better results; however, the difference was minimal and insufficient to justify the use of a larger quantity of the enzyme mixture. We also examined the reaction in the presence of 0.1 M buffer solution (NH4)2SO4 and 0.1 M buffer solution NH4Cl. The results indicated that significantly more glucose was produced in the (NH4)2SO4 buffer solution, and the consumption of NaOH was simultaneously much lower. In the second part of the experiment, we examined the impact of three different temperatures (50, 55 and 60 °C) on the reaction and the enzyme mixture. The data showed that the optimal temperature was 50 °C, as the highest amount of glucose was produced in this reactor.
