Title:Optimizacija in validacija metode za določanje celokupnega dušika v vzorcih tal v odvisnosti od založenosti tal : magistrsko delo

Abstract:Dušik je zanimiv element za analizo, ravno zaradi vseh svojih oblik v zemlji; obstajajo namreč rastlinam dostopne oblike dušika v zemlji in njim nedostopne oblike. Le te se lahko v tleh s pomočjo bakterij pretvorijo iz ene oblike v drugo. Dušik je pomemben element za rast rastlin, saj sodeluje pri fotosintezi, če pa ga je v tleh preveč lahko pride do izpiranja oziroma zmanjšanja dostopnosti drugih elementov, zato je pomembno, da je dušika v zemlji prava količina, glede na vrsto kulture, ki jo gojimo. V magistrskem delu smo izvedli optimizacijo in validacijo metode za določevanje celokupnega dušika v vzorcih tal v odvisnosti od založenosti tal, glede na laboratorij Oddelka za agrokemijo in pivovarstvo na Inštitutu za hmeljarstvo in pivovarstvo Slovenije. Za validacijo in optimizacijo same metode smo sledili metodi za določevanje celokupnega dušika po Kjeldahlu, ki je v skladu s standardom ISO 11261. Ugotovili smo, da je optimalna zatehta vzorca med 1 g in 4 g, optimum samega razklopa pa je 75 min pri temperaturni stopnji 6 za katalizator v tableti oziroma med 75 min in 120 min pri temperaturni stopnji 7 za katalizator v prahu. Sledila je validacija metode, pri čemer je bila potrjena ponovljivost, obnovljivost in točnost metode. Ob koncu validacije lahko potrdimo tudi izkoristek, ki je med 98 % in 117 %.

Title:Optimization and validation of the method for determination of total nitrogen in soil samples depending on soil availability
Abstract:Nitrogen is an interesting element to analyze precisely because it occurs in the soil in different forms, i.e. there are forms of nitrogen in the soil that are accessible to plants and forms that are inaccessible to them. Only these can be transformed from one form to another with the help of bacteria in the soil. Nitrogen is an important element for plant growth as it is involved in photosynthesis. However, if there is too much nitrogen in the soil, it can leach out other elements or reduce their availability. It is therefore important that the right amount of nitrogen is present in the soil, depending on the type of crop being grown. In the master's thesis, we performed the optimization and validation of the analytical method for determination of total nitrogen in soil samples as a function of soil stocking density, which is performed in the laboratory of the Department of Agrochemistry and Brewing at the Institute of Hops research and Brewing Slovenia. For the validation and optimization of the method itself, we followed the method for the determination of total nitrogen according to Kjeldahl, which complies with the ISO 11261 standard. We found that the optimum weight of the sample is between 1 g and 4 g, and the optimum of the digestion itself is 75 min at temperature level 6 for the catalyst in a tablet or between 75 min and 120 min at temperature level 7 for the ground catalyst. This was followed by validation of the method, which confirmed the reproducibility, precision and accuracy of the method. At the end of the validation, we can confirm the recovery, which is between 98 % and 117 %.
