Title:Vpliv akutnega pankreatitisa na dinamiko znotrajceličnih kalcijevih oscilacij celic beta trebušne slinavke v mišjem modelu sladkorne bolezni tipa 2 : magistrsko delo

Abstract:Akutni pankreatitis (AP) je akutno vnetje eksokrinega dela trebušne slinavke, ki resno poveča tveganje bolnikov za razvoj z akutnim pankreatitisom povzročene sladkorne bolezni. V sklopu magistrske naloge smo preučevali vpliv AP na dinamiko kalcijevih oscilacij celic beta trebušne slinavke. AP smo inducirali pri mišjih samcih linije NMRI z intraperitonealnim injiciranjem ceruleina. Indukcija AP pri miših je povzročila povečano aktivnost serumske amilaze in izboljšala glukozno toleranco na tešče 90 in 120 min po začetku glukoznega tolerančnega testa, ni pa vplivala na serumsko koncentracijo inzulina na tešče. Funkcionalno delovanje celic beta po indukciji AP smo ovrednotili s pomočjo akutnih tkivnih rezin trebušne slinavke in s konfokalnim snemanjem dinamike znotrajcelične koncentracije kalcija. Indukcija AP je vplivala na koncentracijsko odvisnost aktivnosti celic beta tako, da je krivuljo premaknila v levo. Dodatno smo razvili nov pristop vrednotenja stopnje AP po injiciranju ceruleina, ki omogoča, da lahko ocenimo stopnjo indukcije AP tudi na živih tkivnih rezinah in sočasno merimo funkcionalno aktivnost endokrinih celic otočkov.

Title:Effect of acute pancreatitis on the dynamics of intracellular calcium oscillations of pancreatic beta cells in a mouse model of type 2 diabetes mellitus
Abstract:Acute pancreatitis (AP) is an acute inflammation of the exocrine part of the pancreas that seriously increases the risk of patients developing AP-induced diabetes. In the context of this MSc thesis, we investigated the impact of AP on the dynamics of pancreatic beta-cell calcium oscillations. AP was induced in male mice of the NMRI line by intraperitoneal injection of cerulein. Induction of AP in mice resulted in increased serum amylase activity and improved fasting glucose tolerance, 90 and 120 min after initiation of the glucose tolerance test, but had no effect on fasting serum insulin concentration. Beta-cell function after AP induction was evaluated using acute pancreatic tissue sections and confocal recording of intracellular calcium concentration dynamics. AP induction affected the concentration dependence of beta cell activity by shifting the curve to the left. Additionally, we developed a novel approach to evaluate the degree of AP after cerulein injection, which allows us to assess the degree of AP induction also in living tissue slices and to simultaneously measure the functional activity of islet endocrine cells.
