Title:Optimizacija sinteze nanoceluloze iz bakterije Komagataeibacter hansenii : diplomsko delo visokošolskega strokovnega študijskega programa I. stopnje

Abstract:Diplomska naloga proučuje vpliv različnih parametrov na sintezo bakterijske celuloze (BC) iz bakterije Komagataeibacter hansenii. V časovnem obdobju šestih tednov smo sintetizirali BC na produkcijskih gojiščih, ki so bila izpostavljena različnim pogojem gojenja. Optimizirali smo naslednje pogoje gojenja: volumen inokuluma, različna produkcijska gojišča z različno koncentracijo vira ogljika, vpliv prisotnosti svetlobe in čas gojenja. BC smo izolirali po postopku čiščenja z NaOH s pomočjo centrifugiranja. Karakterizacija in čistost produkta sta bili določeni s FTIR analizo. Rezultati so pokazali, da je sinteza BC najučinkovitejša pri večjem volumnu dodanega inokuluma, višji koncentraciji vira ogljika v produkcijskem mediju in brez prisotnosti svetlobe. Po štirih tednih gojenja smo zaradi upočasnitve rasti produkta dodali sveže pripravljen gojitveni medij in s tem povečali produkcijo BC v 6 tednih gojenja.

Title:Optimization of nanocellulose synthesis from bacteria Komagataeibacter hansenii
Abstract:The diploma thesis examines the influence of different parameters on the synthesis of bacterial cellulose (BC) from Komagataeibacter hansenii bacteria. Over a period of six weeks, we synthesized BC on production media that were set up under different growing conditions. We optimized the following growing conditions: inoculum volume, different production media with different carbon source concentrations, the influence of light presence and growing time. BC was isolated by purification procedure with NaOH by centrifugation. Product characterization and purity were determined by FTIR analysis. The results showed that BC synthesis is most efficient at higher volume of added inoculum, higher concentration of carbon source in the production medium and in the absence of light. After four weeks of cultivation, due to the slow growth of the products, we added freshly prepared culture medium and thus increased BC production in 6 weeks of cultivation.
