Title:Antioksidativna aktivnost nekaterih vrst in medvrstnih križancev bezga v modelni celični liniji TLT z metodo uporabe barvila DCFH-DA.

Abstract:Antioksidativne snovi pozitivno vplivajo na celice telesa, saj izboljšujejo njihov fitnes in tako pripomorejo k boljšemu zdravju ljudi. Namen te raziskave je bil dokumentirati antioksidativne lastnosti ekstraktov bezgovih jagod. V raziskavo je bilo vključenih 9 različnih genotipov bezga (vrsti Sambucus javanica in S. nigra ter 7 medvrstnih križancev). Antioksidativno aktivnost ekstraktov bezgov smo izmerili s pomočjo uporabe barvila DCFH-DA (2´,7´-diklorofluorescin-diacetat) v pogojih in vitro na modelni celični makrofagni liniji TLT. Na različne načine smo ovrednotili rezultate fluorescence, saj ni določenega enotnega načina za izračun znotrajceličnega antioksidativnega potenciala v primerjavi s pozitivno kontrolo ali standardnimi antioksidanti. Ugotovili smo, da imajo proučevani genotipi bezgov različni znotrajcelični antioksidativni potencial. Najvišji znotrajcelični antioksidativni potencial ima vrsta Sambucus nigra (NI), najnižjega pa medvrstni križanec (JA × NI) × MIQ. Rezultati bi lahko bili zelo koristni za nadaljnjo selekcijo in vzgojo križancev bezga.

Title:Antioxidant Potential of Some Species and Interspecific Hybrids of Elderberries Studied in a TLT Model Cell Line Based on DCFH-DA Staining
Abstract:Antioxidant substances have positive effects on body cells by improving their fitness and hence they contribute to better human health. The purpose of this study was to document the antioxidant properties of elderberry fruit extracts. The study included 9 different genotypes of elderberries (the species Sambucus javanica and S. nigra, and seven interspecific hybrids). The antioxidative activity of elderberry extracts was measured using DCFH-DA staining (2′, 7′-dichlorofluorescin diacetate) in vitro using a model macrophage cell line TLT. Fluorescence results were evaluated in different ways, since there was no specific way to calculate the intracellular antioxidant potential compared to the positive control or standard antioxidants. We found that fruits of the studied genotypes of elderberries had different intracellular antioxidant potential. The highest intracellular antioxidative potential was exhibited by Sambucus nigra (NI) and the lowest by the interspecific hybrid (JA × NI) × MIQ. The results can be very useful for further selection and breeding of elderberry hybrids.
