Title:Prenos divjih genotipov hmelja (Humulus lupulus L.) iz in vitro v in vivo razmere ter njihov odziv na sušo v lončnem poskusu

Abstract:Na Inštitutu za hmeljarstvo in pivovarstvo Slovenije v Žalcu (IHPS) smo v letu 2014 prenesli divje genotipe hmelja (Humulus lupulus L.) iz in vitro v in vivo razmere, ter ugotavljali njihov odziv na sušo v lončnem poskusu. Analizirali in primerjali smo odziv divjega hmelja, nabranega v Sečoveljskih dolinah in sorte Savinjski golding (SG) na sušne razmere. Delo je potekalo v več fazah. V tkivni kulturi smo uporabili genotipe z oznakami SOL 9/11-10, SOL 13-2, SOL 13-9, SOL 9/11-18, SOL 11/11-33 in sorto Savinjski golding. V prvi fazi smo le-te prenesli na gojišče za koreninjenje. Nato smo z metodo aklimatizacije utrdili rastline navedenih genotipov za namen lončnega poskusa. V tretji fazi, ko so bile rastline presajene v lonce, smo pet loncev vsakega genotipa izpostavili sušnim razmeram, medtem ko smo jih pet optimalno zalivali. Po štirinajstih dneh smo naredili vzorčenje listov in po uveljavljenem protokolu določili relativno vsebnost vode (ang. RWC - relative water content). Spremljali smo morfološke spremembe listov in merili vsebnosti vode v njih. Zapisovali, opisovali in fotografirali smo sprotna opažanja. Pridobljene podatke smo uredili in analizirali.

Title:The transfer of wild hop genotypes (Humulus lupulus L.) from in vitro to in vivo conditions and their response on drought in a pot experiment
Abstract:In 2014 we transferred wild genotypes of hop (Humulus lupulus L.) from in vitro to in vivo conditions at the Slovenian Institute of Hop Research and Brewing in Žalec (IHPS) and observed their response on drought in a pot experiment. We analysed and compared the response of wild hops, gathered in Sečovlje Valley as well as the variety Savinjski Golding (SG) on drought. The work was carried out in different steps. In tissue culture, we used genotypes with the labels SOL 9/11-10, SOL 13-2, SOL 13-9, SOL 9/11-18, SOL 11/11-33 and the variety Savinjski Golding. In the first step, we transferred them in the rooting medium. In the next step, the plants were acclimatized and hardened for the purpose of the pot experiment. In the third step, when the plants were transplanted into pots, five pots were exposed to dry conditions, while the others were optimally watered. After fourteen days, we sampled leaves according to the established protocol in order to determine the relative water content (RWC). We monitored the morphological changes of leaves and measured the water content. We regularly made notes, took pictures and collected the data which were later systematically analysed.
