Title:PRIMERJAVA UČINKOVITOSTI RAZLIČNIH MAKROZAMREŽENIH POLIMERNIH XAD ADSORBENTOV PRI ADSORPCIJI VANKOMICINA

Abstract:Vankomicin je antibiotik, ki se uporablja v preventivi ter za zdravljenje okužb z bakterijami. Iz več razlogov velja za zdravilo zadnje izbire. Eden izmed njih je tudi je tudi ta, da so pri prvih preskušanjih zdravila uporabljali nezadostno prečiščeno učinkovino. To pa se je pri pacientih odrazilo v obliki škodljivega vpliva na sluh in ledvica. Vankomicin pridobivamo s fermentacijskim postopkom. Sledi proces izolacije, s katerim odstranimo nečistote kot so barvila in anorganske soli. Izolacijski proces izvajamo v več stopnjah, ena od njih je adsorpcija na makro zamreženi polimerni adsorbent Amberlite XAD 16N, ki je kot stacionarna faza vgrajen v adsorpcijske kolone. Ob vgradnji dodatne adsorpcijske kolone, ki je konstrukcijsko enaka obstoječim napravam, smo adsorpcijsko kolono napolnili s standardno stacionarno fazo makro zamreženim polimernim adsorbentom Amberlite XAD 16N proizvajalca Rohm and Haas. Adsorbent smo pripravili za adsorpcijo po enakem postopku kot v obstoječih adsorpcijskih kolonah. Temu pa je sledil poskusen proces adsorpcije aktivne farmacevtske učinkovine, odstranitev nečistoč in desorpcija prve. Vendar pa z nadaljnjimi stopnjami odstranjevanja nečistoč nismo dosegli zahtevane kvalitete aktivne farmacevtske učinkovine. Cilj tega diplomskega dela je raziskati vpliv sveže vstavljenega adsorbenta Amberlite XAD 16N (proizvajalca Rohm and Haas), ter le tega primerjati s podobnimi makro zamreženim polimernimi adsorbenti. Na laboratorijskem nivoju smo z medsebojno primerjavo želeli ugotoviti, kateri adsorbent je najbolj učinkovit pri izolaciji aktivne farmacevtske učinkovine. V primeru, da se z medsebojno primerjavo ne zazna signifikantnih razlik med posameznimi polimernim adsorbenti, bo potrebno za zagotavljanje ustrezne kakovosti aktivne farmacevtske učinkovine glede na sedanji proces izolacije uvesti dodatno odstranjevanje nečistoč.

Title:COMPARISON OF EFFECTIVENESS FOR DIFFERENT MACRO-CROSSLINKED POLYMERIC XAD ADSORBENTS FOR ADSORBTION OF VANCOMYCIN
Abstract:Vankomicin is antibiotic, which is used as a preventive and also as a treatment of bacterial infections. Due to many reasons, it is so called drug of the last choice.. One of these reasons is the usage of insufficient purified substances in the early phases of testing of the Vancomycin, which reflected in harmful side effects to the patients hearing and kidneys. Vancomycin is produced by a fermentation procedure followed by an isolation process, which removes impurities like dyes and salts. One of the removing levels of impurities within the isolation process, is adsorption onto macro cross-linked polymeric adsorbent Amberlite XAD 16N. After the installation of additional adsorption column, which is structurally identical to the other already existing devices, the new column was filled with the Rohm and Haas (producer) standard stationary phase of macro cross-linked polymeric adsorbent Amberlite XAD 16N. With this type of adsorbent, the isolation of active ingredients is implemented on other existing devices. The procedure of preparing the adsorbent for the adsorption was the same like in other devices, followed by the testing process of the adsorption of active pharmaceutical ingredient, the removal of impurities and desorption phase. With the following stages of removing the impurities, we did not achieve the required quality of active pharmaceutical ingredients. The aim of this thesis is therefore investigate to the influence of additional loaded Rohm and Haas absorbent Amberlite XAD 16N and its comparison with the similar macro-crosslinked polymeric adsorbents. We wanted to find out which adsorbent is the most efficient in isolation of the studied pharmaceutical ingredient. If there is no significant differences between polymeric adsorbents, it will be necessary to implement an additional level of removing the impurities, if we want to assure the best quality of the active pharmaceutical ingredient.
