Title:MOLEKULSKA ANALIZA AKCESIJ SLIV (Prunus domestica L.) Z UPORABO MIKROSATELITSKIH MARKERJEV

Abstract:Ohranjanje in varovanje starih tradicionalnih sort in genotipov sliv (Prunus domestica L.) je bistvenega pomena za ohranjanje biodiverzitete ter vzdrževanje rastlinskih genetskih virov. Genetske materiale hranijo genske banke, ki so namenjene za učinkovito in kakovostno ohranjanje akcesij za daljše časovno obdobje. V raziskavo smo vključili 15 akcesij sliv, ki so locirane v genski banki Fakultete za kmetijstvo in biosistemske vede. Namen diplomskega dela je bil ovrednotiti sorodnost med starimi genotipi sliv z uporabo molekulskih markerjev in morebitne duplikate izločiti iz uradne evidence genske banke FKBV. Ugotavljali smo tudi, ali lahko mikrosatelitske lokuse, ki so bili najdeni pri češnjah (Prunus avium L.), zaradi istega botaničnega rodu uporabimo tudi pri slivah. Na šestih mikrosatelitskih lokusih EMPA001, EMPA002, EMPA003, EMPA004, EMPA011 in EMPA029 smo pri 15 preučevanih genotipih sliv skupaj namnožili 46 polimorfnih alelov, v povprečju 7,666 alela na lokus. Genetske razdalje analiziranih akcesij smo izračunali z Diceovim koeficientom in izdelali dendrogram. Mikrosatelitske lokuse, najdene pri češnjah, smo uspešno uporabili za analize pri slivah. Med 15 analiziranimi akcesijami nismo našli duplikatov.

Title:MOLECULAR ANALYSIS OF PLUMS ACCESSIONS (Prunus domestica L.) USING MICROSATELLITE MARKERS
Abstract:The preservation and protection of traditional species and genotypes of plums (Prunus domestica L.) is essential for the conservation of biodiversity and the maintenance of genetic plant resources. Gene banks store genetic materials with the purpose of efficient and good quality accession maintenance for an extended period of time. We included 15 accessions of plums in our genetic analysis which are located in the Gene bank of the Faculty of Agriculture and Life Sciences. The purpose of the thesis was to evaluate the congeniality between the old plum genotypes based on molecular analysis and to remove any reoccurring copies from the official database of the gene bank. We were also observing whether the Microsatellite loci that were found on Cherries (Prunus avium L.) could be used on plums (P. domestica) due to the same botanical genus. Six microsatellite loci (EMPA001, EMPA002, EMPA003, EMPA004, EMPA011 and EMPA029) were taken into account, 15 studied genotypes were examined and 46 polymorphic alleles were exhibited with an average of 7,666 alleles per loci. The genetic distances between studied accessions were calculated using the Dice coefficient to form a dendrogram. The microsatellite loci found in cherries were successfully applied to plum analysis. There were also no copies found among the 15 analysed accessions.
