Title:DOLOČANJE IZBRANIH SPOJIN V HIDROLIZATU BIOMASE S TEKOČINSKO KROMATOGRAFIJO

Abstract:Namen diplomske naloge je razviti metodo tekočinske kromatografije visoke ločljivosti (angl. high-performance liquid chromatography, HPLC) za določanje izbranih organskih kislin, aldehidov in ketonov v hidrolizatu biomase. Pri hidrolizi biomase nastajajo produkti, ki vsebujejo različne koncentracije organskih kislin, aldehidov, ketonov in sladkorjev. V okviru eksperimenta smo določali DL-gliceraldehid, glikolaldehid, 1,3-dihidroksiaceton (1,3-DHA), metanojsko (mravljično) kislino, etanojsko (ocetno) kislino, levulinsko kislino, 5-hidroksimetilfurfural (5-HMF) in furfural v hidrolizatih olivnih tropin, ki so bile procesirane s subkritično vodo, pri pogojih pod kritično točko vode. Analizirali smo dva vzorca, enega z dodatkom katalizatorja (žveplove kisline) in enega brez dodatka katalizatorja pri podobnem tlaku in temperaturi. Dobljena vzorca smo analizirali z optimirano HPLC metodo s kolono Varian MetaCarb 87H, komponente pa detektirali z UV/VIS detektorjem. Razvito metodo smo validirali in s primerjavo retencijskih časov določili katere od izbranih organskih kislin, aldehidov in ketonov so prisotne v kataliziranem in katere v nekataliziranem vzorcu. Analize so pokazale, da je bilo več preiskovanih komponent v kataliziranem vzorcu, vendar so bile vsebnosti nekaterih komponent v nekataliziranem vzorcu večje.

Title:DETERMINATION OF SELECTED COMPOUNDS IN THE BIOMASS HIDROLYZATE USING LIQUID CHROMATOGRAPHY
Abstract:The purpose of diploma thesis is to develop a high-performance liquid chromatography (HPLC) method for the determination of selected organic acids, aldehydes and ketones in the biomass hydrolyzate. At the hydrolysis of biomass products are being formed that contain different concentrations of organic acids, aldehydes, ketones and sugars. In the experiment, we determined the DL-glyceraldehyde, glycolaldehyde, 1,3-dihydroxyacetone (1,3-DHA), methanoic (formic) acid, etanoic (acetic) acid, levulinic acid, 5-hydroxymethylfurfural (5-HMF) and furfural in the hydrolysates of olive pomace, which had been processed with sub-critical water at conditions below the critical point of water. We analysed two samples; one with the addition of a catalyst (sulfuric acid) and other without the addition of the catalyst at similar pressure and temperature. The obtained samples were analyzed with an optimized HPLC method on a column Varian MetaCarb 87H and components were detected using UV/VIS detector. We validated the developed method and determined which of the selected organic acids, aldehydes and ketones are present in the catalysed and which in the non-catalysed sample. Analyses showed that there were more investigated components in the catalyzed sample, but an amounts of certain components were higher in the non-catalysed sample.
