Title:VPLIV STOPNJE ZRELOSTI NA VSEBNOST SKUPNIH FENOLOV V IZBRANIH SORTAH SLIV (Prunus domestica L.)

Abstract:V diplomskem delu smo spremljali vsebnost skupnih fenolov v času zorenja različnih sort sliv ('Čačanska lepotica', 'Hanita', 'Stanley', 'Tophit' in 'Valor'). Vzorčenje je potekalo v Sadjarskem centru Maribor - Gačnik v letu 2007. Povprečen vzorec je predstavljalo 50 naključno izbranih plodov odvzetih v 2-3 dnevnih presledkih. Plodove smo razkoščičili, homogenizirali in do ekstrakcije hranili v zamrzovalni omari. Vzorce smo ekstrahirali z vodno raztopino acetona (aceton : voda, 70 : 30, v/v) in v ekstraktih določili vsebnost skupnih fenolov spektrofotometrično s Folin – Ciocalteau metodo. V času optimalne zrelosti je bila najvišja vsebnost skupnih fenolov izmerjena pri sorti 'Valor' (127 mg ekvivalentov galne kisline izražene kot mg GAE/100 g sveže mase (SM)), sledile so sorte 'Tophit' (104 mg GAE/100 g SM), 'Čačanska lepotica' (83,5 mg GAE/100 g SM) in 'Stanley' (39,0 mg GAE/100 g SM). Med dozorevanjem 'Čačanske lepotice' se vsebnost skupnih fenolov ni značilno spremenila, medtem ko smo pri sortah 'Valor' in 'Hanita' izmerili statistično pomembno zvišanje vsebnosti skupnih fenolov pri zadnjem terminu vzorčenja. 'Tophit' je pokazal najvišjo vsebnost skupnih fenolov pri optimalni zrelosti. Izmerjena vrednost je bila značilno različna samo od vrednosti določene v prvem terminu vzorčenja. 'Stanley' je bila edina sorta, ki je imela najnižjo izmerjeno vrednost skupnih fenolov pri optimalni zrelosti. Razlike v vsebnosti skupnih fenolov lahko glede na to, da so bile vse sorte pridelane na podoben način in vzorčene na isti lokaciji, pripišemo genetski variabilnosti.

Title:The effect of ripening stage on total phenolics content in selected plum cultivars (Prunus domestica L.)
Abstract:The aim of graduation thesis was to determine the total phenolics content in fruits of different plum cultivars ('Čačanska lepotica', 'Hanita', 'Stanley', 'Tophit' and 'Valor') during the ripening period. Sampling was performed in the Fruit Growing Centre, Gačnik-Maribor in 2007. Plums were harvested for each cultivar at 2-3 day intervals and the average sample was obtained by mixing 50 randomly picked fruits. Samples were extracted using aqueous acetone solution (acetone : water, 70 : 30, v/v). Total phenolics in plum extracts were determined spectrophotometrically according to Folin-Ciocalteu method. At the commercial ripeness stage the highest total phenolic content was recorded in cultivar 'Valor' (127 mg of gallic acid equivalent (GAE)/100 g of fresh weight (FW)), followed by cultivars 'Tophit' (104 mg GAE/100 g FW), 'Čačanska lepotica' (83,5 mg GAE/100 g FW) and Stanley (39,0 mg GAE/100 g FW). During the fruit ripening period, no statistically significant differences in the content of total phenolics were observed in the cultivar 'Čačanska lepotica', whilst in cultivars 'Valor' and 'Hanita' the content detected at the last sampling date was significantly higher to the values determined on the previous sampling dates. 'Tophit' exhibited the highest total phenolics content at the commercial ripeness stage and the obtained value was statistically significant different only to the value determined on the first sampling date. Among the cultivars studied, 'Stanley' was the only cultivar with the lowest value measured at the commercial ripeness stage. Regarding the fact that plum cultivars were grown under the same horticultural practices at the same location, the established differences in total phenolics could be ascribed to genetic variability.
