Title:Vpliv različnih načinov razkuževanja na regeneracijo brstov sliv 'Mirabolana' (Prunus cerasifera Ehrh.)

Abstract:Rastline razmnožujemo z mikropropagacijo, kadar želimo skrajšati čas razmnoževanja in vzgojiti veliko število rastlin na majhnem prostoru, neodvisno od letnega časa, ali kadar je klasično razmnoževanje dolgotrajno in težavno. Namen raziskave je bil razdeljen na dva dela. V prvem delu smo uporabili različne postopke razkuževanja brstov sliv 'Mirabolana' (Prunus cerasifera Ehrh.). Najprej smo vse razkuževali 40 sekund v 70 % etanolu in nato določeno število brstov 15 minut v DICA, 10 minut v DICA ali 15 minut v natrijevem hipokloritu. Najboljši način sterilizacije brstov smo dosegli z 10 minutnim razkuževanjem DICA (45% sterinih brstov). V drugem delu pa je bil naš namen najti čim bolj optimalno gojišče za razmnoževanje. Uporabili smo štiri različna gojišča G1, G2, G3 in G4. Vsako gojišče se je razlikovalo po tem, ali je vsebovalo saharozo ali glukozo, MS (Murashige in Skoog, 1962) ali McCown woody plant medium (Lloyd in McCown 1980) in različne kombinacije rastnih regulatorjev BAP, IBA in NAA. Kot najboljše se je izkazalo gojišče G2 na bazi McCown woody plant medium, kjer smo po treh mesecih dosegli namnožitveni faktor 22,25.

Title:Effect of disinfection on the regeneration of the cherry plum buds (Prunus cerasifera Ehrh.)
Abstract:We reproduce plants with micropropagation when we want to reduce the time of propagation and raise a large number of plants in a small place, independently of the season, or when the classic propagation is long and difficult. The aim of the study was divided into two parts. In the first part we used different disinfection procedures of the cherry plum buds (Prunus cerasifera Ehrh.). First we decontaminated all buds 40 seconds in 70 % ethanol and then the certain number of buds in DICA 15 minutes or 10 minutes and 15 minutes in the Na-hypochlorite. The best results were achieved by the treatment with 10 minutes of DICA, where 45 % of sterile buds were obtained. In the second part it was our intention to find the most optimal medium for propagation. We used four different medium G1, G2, G3 and G4. Each medium were distinguished, whether contained a sucrose or glucose, MS (Murashige and Skoog 1962) or McCown woody plant medium (Lloyd and McCown 1980) and different combinations of growth regulators BAP, IBA, NAA. The medium G2 (based on McCown woody plant medium) proved to be the most successful for in vitro propagation of cherry plum. On this medium the multiplication factor of 22.25 in three months were obtained.
