Title:Časovno spremljanje vsebnosti fotosinteznih pigmentov pri vzgoji potaknjencev mnogocvetnih pelargonij

Abstract:Potaknjenci so po odstranitvi od matične rastline zaradi slabšega privzema vode in hranilnih snovi podvrženi fiziološkemu stresu, ki se odraža v manjši prevodnosti listnih rež, nižjem vodnem potencialu in zmanjšani vsebnosti klorofila. Glavne težave pri razmnoževanju s potaknjenci so slabša sposobnost koreninjenja in senescenca listov. V diplomskem delu nas je zanimal vpliv izvlečka rjavih alg Ecklonia maxima (Kelpak, KelpProduct) na fotosintezne pigmente v listih potaknjencev visečih pelargonij (Pelargonium peltatum (L.) L'Hérit ex Aiton 'Ville de Paris Red') od podtikanja rastlin do faze tržne zrelosti, ki smo jih določili s spektrofotometrično in HPLC metodo. Med poskusom smo potaknjencem talno aplicirali 0,5 %, 1 % in 2 % vodno raztopino Kelpaka v dveh terminih: pred potikanjem v gojitvene plošče ter pred sajenjem v lončke. Liste potaknjencev smo vzorčili štirikrat. Z biokemičnimi analizami smo določili signifikantno povišanje vsebnosti klorofila a (za 166 %) in klorofila b (za 79 %) v tretjem vzorčnem terminu pri tretiranju pelargonij z 2 % koncentracijo Kelpaka. Na akumulacijo luteina v listih sta značilno vplivali 1 % in 2 % vodna raztopina Kelpaka pri vzorcih tretjega in četrtega termina. Vsebnost -karotena se je v pelargonijah znatno povišala v tretjem terminu pri vseh dodanih koncentracijah Kelpaka ter v četrtem terminu pri 2 % koncentraciji Kelpaka. Iz rezultatov je razvidno, da se signifikantno povišane vsebnosti fotosinteznih pigmentov odražajo predvsem v tretjem vzorčnem terminu, pri dodani 1 % in 2 % koncentraciji Kelpaka kot tudi ob fazi tržne zrelosti pelargonij, kar potrjuje dolgoročni učinek Kelpaka na rast in razvoj potaknjencev.

Title:Time-course analysis of photosynthetic pigments during growth and development of cuttings of ivy-leaved pelargonium
Abstract:Due to injuries and severance from the stock plant, cuttings must survive physiological stress, with little water or nutrient uptake. Lower water potential is associated with reduced stomatal conductance and decreased chlorophyll concentrations. Thus, the main problems in propagation of cuttings are leaf senescence and impaired root formation. In the presented experiment, the influence of the brown algae extract (Ecklonia maxima, Kelpak®, KelpProduct) on the photosynthetic pigment contents was studied in leaves of ivy-leaved pelargonium cuttings (Pelargonium peltatum (L.) L'Hérit ex Aiton 'Ville de Paris Red') from the moment of removal from the stock plants until the phase of market maturity. Kelpak was applied at different concentrations (0.5 %, 1 %, and 2 %) in two terms: firstly before cutting were transferred to the multiplates, and secondly before cuttings were replanted in 10 cm pots. The leaves of cuttings were sampled four times during the rooting period. We defined a significant increase in chlorophyll a (166 %) and chlorophyll b (79 %) contents in leaves of 2 % Kelpak treated cuttings at the third sampling date. Furthermore, both 1 % and 2 % Kelpak solutions significantly influenced the accumulation of lutein in leaves at the third and fourth sampling date. The concentrations of β-carotene considerably raised at the third sampling date, in leaves of plants treated with all studied Kelpak concentrations. At the fourth sampling date, the concentrations of β-carotene significantly raised when 2 % Kelpak concentration had been applied. The results showed, that photosynthetic pigment contents significantly raised, especially at the third sampling date when cuttings had been treated with 1 % and 2 % Kelpak concentrations, as well as at the phase of market maturity. Our results demonstrated long-term effect of Kelpak on the growth and development of pelargonium cuttings.
