Title:OPTIMIZACIJA PROCESNIH POGOJEV KRISTALIZACIJE FARMACEVTSKE UČINKOVINE

Abstract:Kristalizacija je pomembna kemijska in tehnološka operacija, s katero iz produkta odstranimo nečistoče, zagotovimo pravilno polimorfno obliko ter želeno velikost in obliko delcev. Pri kristalizaciji je pomembna izbira primernega topila, saj je od tega odvisna uspešnost čiščenja. Diplomsko delo predstavlja določitev najprimernejšega topila za kristalizacijo in študijo vplivov procesnih parametrov na potek kristalizacije surovega produkta SIK-a (4'-[(2-Butil-4-okso-1,3-diazaspiro[4,4]non-1-en-3-il)metil]bifenil-2-karbonitril). SIK je ključni intermediat za sintezo zdravilne učinkovine Irbesartan, ta pa se v farmacevtski industriji uporablja kot učinkovina za zdravljenje bolezni srca in ožilja, centralnega živčevja, glavkoma, retinopatije ter diabetične nefropatije. Z določitvijo najprimernejšega topila za kristalizacijo in procesnih parametrov, smo na koncu izolirali produkt SIK, ki je ustrezal vsem zahtevam (oblika, čistota, izkoristek, IR, NMR, EA).

Title:OPTIMIZATION OF PROCESS PARAMETERS FOR CRYSTALIZATION OF ACTIVE PHARMACEUTICAL INGREDIENTS
Abstract:Crystallization is an important chemical and technological operation through which we eliminate impurity from the product, assure the correct polymorph shape and the length and shape of parts. During crystallization the choice of the appropriate solvent is very important because the efficiency of the cleaning depends on it. Thesis defines the most suitable solvent for crystallization and the study of the influences that the process parameters have on the course of crystallization of the final raw product SIK (4'-[(2-Butyl-4-oxo-1,3-diazaspiro[4.4]non-1-en-3-yl)methyl]biphenyl-2-carbonitrile). SIK is a key pharmaceutical intermediate for the synthesis of the active substances of Irbesartan, which is in pharmaceutical industry used as a substance for treating heart and vascular diseases, central nervous system, glaucoma, retinopathy and diabetic nephropathy. With defining the most suitable solvent for crystallization and process parameters we at the end isolated product SIK that suited all of set specifications (shape, cleanliness, usage, IR, NMR, EA).
