Saviour siblings - current overview, dilemmas and possible solutions?

Abstract: In certain cases, when an already born child is affected with a fatal disease and needs transplantation, the only possible solution could be the creation of saviour sibling. Saviour sibling is a term that refers to a child born through a procedure in which an embryo, created in vitro, is being tested in order to determine whether such an embryo could provide stem cells or tissue for an already born, ill child. If the embryo is both, a matching donor and free of the disease, it could be implanted and after the birth of a child, the umbilical cord stem cells or tissue could be used for treatment of a sick sibling. However, this procedure poses a number of dilemmas. This paper aims to give a brief analysis of these issues, to address some of the main concerns and to provide possible solutions for future regulation of this technique.

Rules on assisted reproduction on Slovenia - collision between legislation on biomedically assisted procreation and regulation relating to safety and quality of tissues and cells

Abstract: Regulatory landscape on assisted reproduction (AR) has changed over the last 20 years in Slovenia. Notwithstanding the fact that in-field legislation (Infertility Treatment and Procedures of Biomedically Assisted Procreation Act - ZZNPOB) has not been amended or derogated since its adoption in 2000, the regulatory framework has been supplemented with legislative acts, implementing EU Directives on safety and quality of tissues and cells used in treatment. Implementation of the latter (also through adoption of Act on Quality and Safety of Human Tissues and Cells for the Purposes of Medical Treatment - ZKVČTC in 2007) has predominantly failed to acknowledge the specifics of assisted reproduction (regulated by ZZNPOB), resulting in regulatory framework which lacks legal certainty and predictability. Article therefore addresses the collision between ZZNPOB and ZKVČTC and proposes solutions de lege ferenda to avoid shortcomings of existing regulation.

Critical and supercritical spatiotemporal calcium dynamics in beta cells

Abstract: A coordinated functioning of beta cells within pancreatic islets is mediated by oscillatory membrane depolarization and subsequent changes in cytoplasmic calcium concentration. While gap junctions allow for intraislet information exchange, beta cells within islets form complex syncytia that are intrinsically nonlinear and highly heterogeneous. To study spatiotemporal calcium dynamics within these syncytia, we make use of computational modeling and confocal high-speed functional multicellular imaging. We show that model predictions are in good agreement with experimental data, especially if a high degree of heterogeneity in the intercellular coupling term is assumed. In particular, during the first few minutes after stimulation, the probability distribution of calcium wave sizes is characterized by a power law, thus indicating critical behavior. After this period, the dynamics changes qualitatively such that the number of global intercellular calcium events increases to the point where the behavior becomes supercritical. To better mimic normal in vivo conditions, we compare the described behavior during supraphysiological non-oscillatory stimulation with the behavior during exposure to a slightly lower and oscillatory glucose challenge. In the case of this protocol, we observe only critical behavior in both experiment and model. Our results indicate that the loss of oscillatory changes, along with the rise in plasma glucose observed in diabetes, could be associated with a switch to supercritical calcium dynamics and loss of beta cell functionality.

Does the development of alternative energy technologies allow for new forms of coopetition?

Abstract: The article at hand illustrates how new types of coopetition emerge in the transformation of large technical systems. It builds on the latest literature on coopetition and highlights diverse institutional arrangements for coopetition, their effects on the actual innovation and the potential benefits for the firms involved. In contrast to many incremental innovations, the transformation of large technical systems requires the cooperation of many diverse actors as various resources are needed. This does not only open up the opportunity of new private-private or public-private cooperations but also brings about various new forms of commonly performed practices.

Use of non-conventional cell disruption method for extraction of proteins from black yeasts

Abstract: The influence of pressure and treatment time on cells disruption of different black yeasts and on activities of extracted proteins using supercritical carbon dioxide process was studied. The cells of three different black yeasts Phaeotheca triangularis, Trimatostroma salinum, and Wallemia ichthyophaga were exposed to supercritical carbon dioxide (SC $CO_2$) by varying pressure at fixed temperature (35°C). The black yeasts cell walls were disrupted, and the content of the cells was spilled into the liquid medium. The impact of SC $CO_2$ conditions on secretion of enzymes and proteins from black yeast cells suspension was studied. The residual activity of the enzymes cellulase, $beta$-glucosidase, $alpha$-amylase, and protease was studied by enzymatic assay. The viability of black yeast cells was determined by measuring the optical density of the cell suspension at 600 nm. The total protein concentration in the suspension was determined on UV–Vis spectrophotometer at 595 nm. The release of intracellular and extracellular products from black yeast cells was achieved. Also, the observation by an environmental scanning electron microscopy shows major morphological changes with SC $CO_2$-treated cells. The advantages of the proposed method are in a simple use, which is also possible for heat-sensitive materials on one hand and on the other hand integration of the extraction of enzymes and their use in biocatalytical reactions.

Quality and delivery of milk in Slovenia

Abstract: Milk production represents the possibility of earning monthly income and assures social security to many farms in Slovenia. In Slovenia the European rules and regulations on the quality of the delivered milk have been introduced, relating to the hygienic quality of milk and the count of somatic cells. The purchase price of the milk depends also on the protein and fat content. The aim of the research was to establish, whether the quality of the delivered milk complies with the prescriptions regulated by the Slovene legislation. The quality of the delivered milk in the area of the Šentjur agricultural cooperative (ACŠ) and in the entire area of Slovenia (SLO) has been studied. The bacteriological data in the individual quality classes, the data on the total count of somatic cels and the data on the fat and protein contents have been analyzed. 36 monthly accounts have been included in statistical processing. It has been established that the delivered milk of the areas in question does not differ in the relevant quality values.

Monitoring the evaporation of fluids from fiber-optic micro-cell cavities

Abstract: Fiber-optic sensors provide remote access, are readily embedded within structures, and can operate in harsh environments. Nevertheless, fiber-optic sensing of liquids has been largely restricted to measurements of refractive index and absorption spectroscopy. The temporal dynamics of fluid evaporation have potential applications in monitoring the quality of water, identificationof fuel dilutions, mobile point-of-care diagnostics, climatography and more. In this work, the fiber-optic monitoring of fluids evaporation is proposed and demonstrated. Sub-nano-liter volumes of a liquid are applied to inline fiber-optic micro-cavities. As the liquid evaporates, light is refracted out of the cavity at the receding index boundary between the fluid and the ambient surroundings. A sharp transient attenuation in the transmission of light through the cavity, by as much as 50 dB and on a sub-second time scale, is observed. Numerical models for the transmission dynamics in terms of ray-tracing and wavefront propagation are provided. Experiments show that the temporal transmission profile can distinguish between different liquids.

The analysis of intracellular and intercellular calcium signaling in human anterior lens capsule epithelial cells with regard to different types and stages of the cataract

Abstract: In this work we investigated how modifications of the Ca2+ homeostasis in anterior lens epithelial cells (LECs) are associated with different types of cataract (cortical or nuclear) and how the progression of the cataract (mild or moderate) affects the Ca2+ signaling. We systematically analyzed different aspects of intra- and inter-cellular Ca2+ signaling in the human LECs, which are attached to surgically isolated lens capsule (LC), obtained during cataract surgery. We monitored the temporal and spatial changes in intracellular Ca2+ concentration after stimulation with acetylcholine by means of Fura-2 fluorescence captured with an inverted microscope. In our analysis we compared the features of Ca2+ signals in individual cells, synchronized activations, spatio-temporal grouping and the nature of intercellular communication between LECs. The latter was assessed by using the methodologies of the complex network theory. Our results point out that at the level of individual cells there are no significant differences when comparing the features of the signals with regard either to the type or the stage of the cataract. On the other hand, noticeable differences are observed at the multicellular level, despite inter-capsule variability. LCs associated with more developed cataracts were found to exhibit a slower collective response to stimulation, a less pronounced spatio-temporal clustering of LECs with similar signaling characteristics. The reconstructed intercellular networks were found to be sparser and more segregated than in LCs associated with mild cataracts. Moreover, we show that spontaneously active LECs often operate in localized groups with quite well aligned Ca2+ activity. The presence of spontaneous activity was also found to affect the stimulated Ca2+ responses of individual cells. Our findings indicate that the cataract progression entails the impairment of intercellular signaling thereby suggesting the functional importance of altered Ca2+ signaling of LECs in cataractogenesis.

The influence of royal jelly and human interferon-alpha (HuIFN-[alpha]N3) on proliferation, glutathione level and lipid peroxidation in human colorectal adenocarcinoma cells in vitro

Abstract: Among royal jelly’s (RJ) various biological activities, its possible antitumour activity deserves particular attention. The purpose of this study was to investigate the influence of RJ, its bioactive component 10-hydroxy-2-decenoic acid (10- HDA), and human interferon-alpha (HuIFN-αN3) on the proliferation of human colorectal adenocarcinoma cells (CaCo- 2), and ascertain their effect on intracellular glutathione (GSH) level and lipid peroxidation. We studied the antiproliferative (AP) activity of RJ [(0.1 g/10 mL phosphate buffer saline (PBS)], HuIFN-αN3 (1000 I.U. mL-1), 10-HDA at 100.0 μmol L-1, and their different combinations, in the ratio 1:1, 1:2, and 2:1 on CaCo-2 cells. The GSH level was measured by glutathione assay. The lipid peroxidation was measured by malondialdehyde (MDA) assay. Single RJ had a low AP activity: 2.0 (0.5 mg mL-1). HuIFN-αN3 had an AP activity of 2.5 (208.33 I.U. mL-1), while 10-HDA had an AP activity of 1.5 (37.5 μmol mL-1). The highest AP activity of 3.8 was obtained when RJ and HuIFN-αN3 were applied at the ratio 2:1. In that combination the level of GSH was 24.9±2.4 nmol g-3 of proteins (vs. 70.2±3.2 nmol g-3 in the control) and the level of MDA was 72.3±3.1 nmol g-3 (vs. 23.6±9.1 nmol g-3 in the control). It is generally assumed that 10-HDA, an important constituent of RJ, together with HuIFN-αN3, is responsible for the inhibition of CaCo-2 cells proliferation in vitro. In our study, however, RJ and HuIFN-αN3 applied at 2:1 decreased the level of GSH the most and significantly increased lipid peroxidation via MDA in CaCo-2 cells. Future studies should show whether these GSH- and MDA-related activities of RJ, HuIFN-αN3, 10-HDA, and their combinations may decrease the tumorigenicity index and tumorigenic potential of various tumour cells in vitro.
