Slow growth storage of Berula erecta in vitro - effect of sucrose, sorbitol and temperature

Opis: It has been established that in vitro storage of Berula erecta (Huds.) Coville by slow growth would rationalize the culture maintenance by reducing the number of transfers to fresh medium. Attempts of slow growth were made while maintaining viability by adding sugars and sugar alcohols to the MS medium and lowering the culture temperature in the growth chambers. The effect of low temperatures (13°C and 4°C), different combinations and concentrations of sorbitol (20, 40 g L-1) and sucrose (30, 40 g L-1), and light was tested to determine the viability and ability of B. erecta to recover growth. Increasing sucrose concentration did not inhibit or slow growth. At 23 ± 2°C, the addition of 20 g L-1 sorbitol successfully inhibited growth, while the addition of 40 g L-1 sorbitol resulted in plant senescence after two months of culturing. Storage of B. erecta at 13°C did not effectively inhibit growth and recovery ability in any treatment. The most effective way to maintain B. erecta for long-term slow growth storage in tissue culture while retaining a high level of viability with minimal growth was to culture the plants at 4°C in the dark. This kept the plants viable and capable of regeneration for several months.

Fran Miklošič in slovenska imena mesecev

Opis: V prispevku je obravnavana jezikoslovna razprava Frana Miklošiča Die slavischen Monatsnamen (Slovanska imena mesecev, 1867), z omejitvijo prikaza slovenskih imen mesecev. Miklošič je zapisal kar 95 različnih slovenskih poimenovanj za mesece. Ta so najpogosteje povezana s poimenovanji za praznike in običaje ter z rastlinstvom, nekoliko manj pa z (običajno kmečkimi) opravili in živalstvom.

Petelinčki Vesne Radovanovič v lahkem branju : magistrsko delo

Opis: Magistrsko delo se ukvarja z bralno pismenostjo učencev četrtega razreda nižjega izobrazbenega standarda. Sestavljeno je iz dveh ključnih delov, teoretičnega in empiričnega dela. Na začetku teoretičnega dela so podrobneje opredeljeni branje in strategije branja, temu sledijo naslednje teme: opredelitev gradnikov bralne pismenosti in natančna razlaga vsakega posameznega gradnika, predstavljeni so tudi model lahkega branja in nekateri njegovi najpomembnejši vidiki. V raziskavi magistrskega dela so sodelovali otroci z intelektualno oviranostjo, zato je eno izmed poglavij v teoretičnem delu namenjeno tudi njim, prav tako tudi prilagojenemu izobraževalnemu programu vzgoje in izobraževanja z nižjim izobrazbenim standardom, ki ga obiskujejo otroci, vključeni v raziskavo. Posebej opredeljen in predstavljen je tudi pojem slikanice, temu se pridružuje tudi predstavitev serije slikanic o petelinčku. Na kratko se teoretični del dotakne tudi avtorice in ilustratorke serije slikanic petelinček. Empirični del zajema in predstavlja celoten načrt izvedbe raziskave magistrskega dela, v njem so predstavljene tudi slikanice serije petelinček, prilagojene po modelu lahkega branja, ki predstavljajo glavni raziskovalni instrument magistrskega dela. Prav tako so predstavljeni tudi rezultati raziskave, natančna in obsežna interpretacija ter analiza teh, ki so pokazali, da so otroci, vključeni v raziskavo, dobro bralno pismeni, ne glede na njihove zmanjšane intelektualne zmožnosti in druge primanjkljaje, ki jih imajo.

Primerjava različnih didaktičnih pristopov na usvajanje lokomotornih spretnosti mlajših otrok : diplomsko delo

Opis: V teoretičnem delu smo najprej predstavili razvoj otroka in nato dejavnike, ki nanj vplivajo. To so dednost, okolje in lastna aktivnost. Predstavili smo gibanje, gibalni razvoj in otrokov gibalni razvoj. Opisali smo temeljne gibalne spretnosti, kamor spadajo pedipulacije oziroma lokomocije, manipulacije in sestavljena gibanja. Vse tri smo podrobneje opisali. Na koncu smo opisali, kaj so didaktični pristopi in dva predstavili, in sicer neposredno usmerjanje otroka in usmerjanje preko igre, ki sta bila ključna v raziskavi diplomske naloge. Otroci so se preko teh dveh didaktičnih pristopov učili lokomotornih spretnosti (tek, prisunski poskoki naprej, poskoki po eni nogi, hopsanje, skok v daljino z mesta in prisunski poskoki v stran). Vse tri skupine otrok (eksperimentalna skupina 1, eksperimentalna skupina 2 in kontrolna skupina) smo posneli, ko so te naloge izvajali, in gibanja otrok ocenili s pomočjo merskega postopka TGMD-3. Snemanje smo ponovili trikrat, saj smo posneli začetno, vmesno in končno stanje. Rezultate smo nato analizirali in jih interpretirali. Glavni cilj diplomske naloge je bil ugotoviti, s katerim didaktičnim pristopom (neposredno usmerjanje ali usmerjanje preko igre) se otroci bolje naučijo ciljnega gibanja in kateri didaktični pristop jih je bolj motiviral. Skozi celotno raziskavo smo prišli do ugotovitve, da se otroci ciljnega gibanja bolje naučijo skozi igro, saj jih le-ta bolj motivira in je zanje bolj zanimiva.

The role of rhetoric in teacher virtual exchanges

Opis: Educators’ rhetorical skills are pivotal in virtual collaboration between teachers and parents, enhancing communication clarity and overcoming cultural barriers, thereby enriching educational experience and fostering intercultural understanding. A review of scientific databases such as Academic Search Complete, ERIC, SCOPUS, and Web of Science, was conducted to identify peer-reviewed articles from 2018 onwards that explore the role of rhetoric in teacher virtual exchanges. Articles were analysed for themes, findings, biases, and research gaps. A notable finding is the lack of research on rhetoric in virtual teacher exchanges, despite its potential to overcome cultural barriers and enhance collaboration. The study reveals that effective rhetorical techniques can improve clarity, persuasiveness, and intercultural understanding in virtual exchanges, indicating a need for more research and educational programs in this area. Furthermore, the study extends to virtual communication between teachers and parents, especially with non-native speakers or those with limited proficiency in the official language. This highlights the need for teachers to use sophisticated rhetorical strategies in diverse linguistic situations to improve understanding and involvement. In summary, the research underscores the significant impact of rhetorical skills in virtual teacher exchanges, with broader implications for global collaboration and intercultural understanding in the educational sphere.

Modeling the amino acid effect on glucagon secretion from pancreatic alpha cells

Opis: Type 2 Diabetes Mellitus (T2DM) is a burdensome problem in modern society, and intensive research is focused on better understanding the underlying cellular mechanisms of hormone secretion for blood glucose regulation. T2DM is a bi-hormonal disease, and in addition to 100 years of increasing knowledge about the importance of insulin, the second hormone glucagon, secreted by pancreatic alpha cells, is becoming increasingly important. We have developed a mathematical model for glucagon secretion that incorporates all major metabolic processes of glucose, fatty acids, and glutamine as the most abundant postprandial amino acid in blood. In addition, we consider cAMP signaling in alpha cells. The model predictions quantitatively estimate the relative importance of specific metabolic and signaling pathways and particularly emphasize the important role of glutamine in promoting glucagon secretion, which is in good agreement with known experimental data.

Diversity and content of carotenoids and other pigments in the transition from the green to the red stage of Haematococcus pluvialis microalgae identified by HPLC-DAD and LC-QTOF-MS

Opis: H. pluvialis is a unicellular freshwater alga containing many bioactive compounds, especially carotenoids, which are the strongest antioxidants among the pigments. This study evaluates the composition and content of carotenoids and other pigments in both stages of algae life cycle, especially in the green vegetative stage, less studied in comparison to the red stage. To determine the composition and content of carotenoids, a combination of HPLC-DAD and LC-QTOF-MS was used. The content of carotenoids in the green vegetative stage was significantly lower than in the red vegetative stage. In the green vegetative stage, 16 different carotenoids and other pigments were identified. Among the total 8.86 mg g−1 DW of pigments, 5.24 mg g−1 DW or 59% of them were chlorophyll a with its derivatives, and 3.62 mg g−1 DW or 41% of them were free carotenoids. After the transition from the green to the red stage, the carotenoid composition was replaced by secondary carotenoids, astaxanthin and its esters, which predominated in the whole carotenoid composition. In addition to free astaxanthin, 12 astaxanthin monoesters, 6 diesters and 13 other carotenoids were determined. The majority of 37.86 mg g−1 DW pigments were monoesters. They represented 82% of all pigments, and their content was about 5 times higher than both, diesters (5.91 mg g−1 DW or 12% of all) and free carotenoids (2.4 mg g−1 DW or 6% of all). The results of the study contribute to the data on the overall pigment composition and content of H. pluvialis algae and provide the basis for further improvement of cultivation of the H. pluvialis algae.

Age-related changes in lipid and glucose levels associated with drug use and mortality : an observational study

Opis: Background: The pathogenesis of type 2 diabetes mellitus is complex and still unclear in some details. The main feature of diabetes mellitus is high serum glucose, and the question arises of whether there are other statistically observable dysregulations in laboratory measurements before the state of hyperglycemia becomes severe. In the present study, we aim to examine glucose and lipid profiles in the context of age, sex, medication use, and mortality. Methods: We conducted an observational study by analyzing laboratory data from 506,083 anonymized laboratory tests from 63,606 different patients performed by a regional laboratory in Slovenia between 2008 and 2019. Laboratory data-based results were evaluated in the context of medication use and mortality. The medication use database contains anonymized records of 1,632,441 patients from 2013 to 2018, and mortality data were obtained for the entire Slovenian population. Results: We show that the highest percentage of the population with elevated glucose levels occurs approximately 20 years later than the highest percentage with lipid dysregulation. Remarkably, two distinct inflection points were observed in these laboratory results. The first inflection point occurs at ages 55 to 59 years, corresponding to the greatest increase in medication use, and the second coincides with the sharp increase in mortality at ages 75 to 79 years. Conclusions: Our results suggest that medications and mortality are important factors affecting population statistics and must be considered when studying metabolic disorders such as dyslipidemia and hyperglycemia using laboratory data.

Perspectives on lessons from the COVID-19 outbreak for post-pandemic higher education: continuance intention model of forced online distance teaching

Opis: The response of most universities to the Coronavirus disease (COVID-19) pandemic was Online Distance Teaching (ODT), which was a new experience for many educators and students. The aim of the study was to investigate the response of university teachers to ODT. A questionnaire was sent to all university teachers (N = 914). We received 290 usable responses. To create a Continuance Intention Model of Forced Online Distance Teaching(CIMoFODT), Confirmatory Factorial Analysis (CFA) and Structural Equation Modelling (SEM) were used in addition to descriptive and inferential statistics. The main findings were as follows: (i) during the closure, use of the videoconferencing system MS Teams was the only item that increased significantly, owing to mandatory use; (ii) the increase in the use of other applications (e.g., Moodle, email) was minimal; (iii) after the reopening of the university, email, Moodle, and supplementary online materials will be used for ODT; MS Teams will be used for small group teaching and individual consultations; (iv) CIMoFODT can be applied to explain the intention to continue ODT. The main conclusion is that teachers will return to traditional teaching when classrooms reopen.

Lipotoxicity in a vicious cycle of pancreatic beta cell exhaustion

Opis: Hyperlipidemia is a common metabolic disorder in modern society and may precede hyperglycemia and diabetes by several years. Exactly how disorders of lipid and glucose metabolism are related is still a mystery in many respects. We analyze the effects of hyperlipidemia, particularly free fatty acids, on pancreatic beta cells and insulin secretion. We have developed a computational model to quantitatively estimate the effects of specific metabolic pathways on insulin secretion and to assess the effects of short- and long-term exposure of beta cells to elevated concentrations of free fatty acids. We show that the major trigger for insulin secretion is the anaplerotic pathway via the phosphoenolpyruvate cycle, which is affected by free fatty acids via uncoupling protein 2 and proton leak and is particularly destructive in long-term chronic exposure to free fatty acids, leading to increased insulin secretion at low blood glucose and inadequate insulin secretion at high blood glucose. This results in beta cells remaining highly active in the “resting” state at low glucose and being unable to respond to anaplerotic signals at high pyruvate levels, as is the case with high blood glucose. The observed fatty-acid-induced disruption of anaplerotic pathways makes sense in the context of the physiological role of insulin as one of the major anabolic hormones.
