Some comments on the legal regulation on misinformation and cyber attacks conducted through online platforms

Opis: What are the tools to effectively manage the new threats to the free flow of opinions to protect this essential precondition for a pluralist social and political system? As a basis for action at the community level, how can we protect users who use internet platforms to inform themselves on issues of public interest from disinformation attacks through cyberspace? Effective action against cyber-attacks that adversely affect certain fundamental rights requires a combination of instruments, creating the technological, economic, human and legal conditions for meaningful counter-measures. In legal terms, the guarantees that platform providers must offer each user to prevent cyber attacks and illegal content should be laid down, and legal instruments shou ld be put in place to ensure they are always available. In addition, in the event of misusing any content shared on the platform or of personal data made available to the operator, clear responsibilities should be established, and the extent to which the responsibility for protection lies with the platform operator or the user should be clarified. In our study, we outline regulatory options to address these challenges.

Dance creativity in kindergarten : improvisation as a method for identifying dance talent in children

Opis: Dance improvisation is a spontaneous and free form of expression that intertwines movement and imagination, creating space for creativity. The creativity expressed through dance improvisation represents a key category in identifying dance talent in children, as it provides insight into their ability for free expression and original movement. The aim of our study was to explore the characteristics of dance improvisation in early childhood, with a focus on physical activity and the formation of movement in children. We employed a quantitative research approach, collecting data through an observational protocol. The study included 246 children aged 5-6 years from 38 public kindergartens across Slovenia. Our findings serve as an important foundation for further research on monitoring dance improvisation in preschool children and for a deeper understanding of the role of body movement in children's creative dance expression.

Mesenchymal stem cells isolated from paediatric paravertebral adipose tissue show strong osteogenic potential

Opis: Mesenchymal stem cells (MSCs) represent the basis of novel clinical concepts in cellular therapy and tissue regeneration. Therefore, the isolation of MSCs from various tissues has become an important endeavour for stem cell biobanking and the development of regenerative therapies. Paravertebral adipose tissue is readily exposed during spinal procedures in children and could be a viable source of stem cells for therapeutic applications. Here, we describe the first case of MSCs isolated from paravertebral adipose tissue (PV-ADMSCs), obtained during a routine spinal surgery on a child. Using quantitative real-time PCR and flow cytometry, we show that PV-ADMSCs have different levels of stem marker expression compared to the MSCs from other sources while having the highest proliferation rate. Furthermore, we evaluate the multipotency of PV-ADMSCs by the three-lineage (adipogenic, osteogenic and chondrogenic) differentiation and compare it to the multipotency of MSCs from other sources. It was found that the PV-ADMSCs have a strong osteogenic potential in particular. Taken together, our data indicate that PV-ADMSCs meet the criteria for successful cell therapy, defined by the International Society for Cellular Therapy (ISCT), and thus, could provide a source of MSCs that is relatively easy to isolate and expand in culture. Due to their strong osteogenic potential, these cells provide a promising basis, especially for orthopaedic applications.

Single-cell transcriptomic and targeted genomic profiling adjusted for inflammation and therapy bias reveal CRTAM and PLCB1 as novel hub genes for anti-tumor necrosis factor alpha therapy response in Crohn’s disease

Opis: The lack of reliable biomarkers in response to anti-TNFα biologicals hinders personalized therapy for Crohn’s disease (CD) patients. The motivation behind our study is to shift the paradigm of anti-TNFα biomarker discovery toward specific immune cell sub-populations using single-cell RNA sequencing and an innovative approach designed to uncover PBMCs gene expression signals, which may be masked due to the treatment or ongoing inflammation; Methods: The singlecell RNA sequencing was performed on PBMC samples from CD patients either naïve to biological therapy, in remission while on adalimumab, or while on ustekinumab but previously non-responsive to adalimumab. Sieves for stringent downstream gene selection consisted of gene ontology and independent cohort genomic profiling. Replication and meta-analyses were performed using publicly available raw RNA sequencing files of sorted immune cells and an association analysis summary. Machine learning, Mendelian randomization, and oligogenic risk score methods were deployed to validate DEGs highly relevant to anti-TNFα therapy response; Results: This study found PLCB1 in CD4+ T cells and CRTAM in double-negative T cells, which met the stringent statistical thresholds throughout the analyses. An additional assessment proved causal inference of both genes in response to anti-TNFα therapy; Conclusions: This study, jointly with an innovative design, uncovered novel candidate genes in the anti-TNFα response landscape of CD, potentially obscured by therapy or inflammation.

Adipose tissue gene expression of entire male, immunocastrated and surgically castrated pigs

Opis: Differences in adipose tissue deposition and properties between pig male sex categories, i.e., entire males (EM), immunocastrates (IC) and surgical castrates (SC) are relatively well-characterized, whereas the underlying molecular mechanisms are still not fully understood. To gain knowledge about the genetic regulation of the differences in adipose tissue deposition, two different approaches were used: RNA-sequencing and candidate gene expression by quantitative PCR. A total of 83 differentially expressed genes were identified between EM and IC, 15 between IC and SC and 48 between EM and SC by RNA-sequencing of the subcutaneous adipose tissue. Comparing EM with IC or SC, upregulated genes related to extracellular matrix dynamics and adipogenesis, and downregulated genes involved in the control of lipid and carbohydrate metabolism were detected. Differential gene expression generally indicated high similarity between IC and SC as opposed to EM, except for several heat shock protein genes that were upregulated in EM and IC compared with SC. The candidate gene expression approach showed that genes involved in lipogenesis were downregulated in EM compared with IC pigs, further confirming RNA-sequencing results.

Peripheral blood transcriptome in breast cancer patients as a source of less invasive immune biomarkers for personalized medicine, and implications for triple negative breast cancer

Opis: Transcriptome studies of peripheral blood cells can advance our understanding of the systemic immune response to the presence of cancer and the mechanisms underlying cancer onset and progression. This enables the identification of novel minimally invasive immune biomarkers for early cancer detection and personalized cancer management and may bring forward new immunotherapy options. Recent blood gene expression analyses in breast cancer (BC) identified distinct patient subtypes that differed in the immune reaction to cancer and were distinct from the clinical BC subtypes, which are categorized based on expression of specific receptors on tumor cells. Introducing new BC subtypes based on peripheral blood gene expression profiles may be appropriate, since it may assist in BC prognosis, the identification of patients likely to benefit from immunotherapy, and treatment efficacy monitoring. Triple-negative breast cancer (TNBC) is an aggressive, heterogeneous, and difficult-to-treat disease, and identification of novel biomarkers for this BC is crucial for clinical decision-making. A few studies have reported TNBC-enriched blood transcriptional signatures, mostly related to strong inflammation and augmentation of altered immune signaling, that can differentiate TNBC from other classical BC subtypes and facilitate diagnosis. Future research is geared toward transitioning from expression signatures in unfractionated blood cells to those in immune cell subpopulations.

Immunocastration in adult boars as a model for late-onset hypogonadism

Opis: Background: While immunocastration has been studied in male pre-pubertal pigs, data on older, sexually mature animals are limited. To understand the physiological effects of androgen deprivation in the late sexual development phase, we compared mature immunocastrated boars (n = 19; average age = 480 days) to young male immunocastrated pigs (n = 6; average age = 183 days) and young entire males (n = 6; average age = 186 days) as positive and negative controls, respectively. Objectives: We hypothesized that the timing of gonadotropin-releasing hormone suppression (early or late sexual development phases) influences the extent of reproductive function inhibition, histological structure of testicular tissue, and expression levels of selected genes related to steroid metabolism. Materials and methods: Antibody titer, hormonal status, and histomorphometric analysis of testicular tissue were subjected to principal component analysis followed by hierarchical clustering to evaluate the immunocastration effectiveness in mature boars. Results: Hierarchical clustering differentiated mature immunocastrated boars clustered with young immunocastrated pigs from those clustered with entire males. Although all mature immunocastrated boars responded to vaccination, as evidenced by the increased gonadotropin-releasing hormone antibody titers (p < 0.001), decreased serum luteinizing hormone concentrations (p = 0.002), and changes in testicular tissue vascularization (lighter and less red testicular parenchyma; p ≤ 0.001), the responses were variable. Sharp decreases in testes index (p < 0.001), Leydig cell volume density (p < 0.001), Leydig cell nucleus-to-cytoplasm ratio (p < 0.001), and testosterone concentration (p < 0.001) were observed in mature immunocastrated boars clustered with young immunocastrated pigs compared with those that clustered with entire males. Additionally, mature immunocastrated boars clustered with young immunocastrated pigs showed lower hydroxysteroid 17-beta dehydrogenase 7 expression than entire males (p < 0.05). The young immunocastrated pigs group showed higher folliclestimulating hormone receptors than the entire males and mature immunocastrated boars, lower steroidogenic acute regulatory protein expression levels compared with entire males, and mature immunocastrated boars clustered with entire males (p < 0.01). Conclusion: The two-dose vaccination regime resulted in progressive but variable regression of testicular function in adult (post-pubertal) pigs; however, it was insufficient to induce a complete immunocastration response in all animals.

New computational models for better predictions of the soil-compression index

Opis: The compression index is one of the important soil parameters that are essential for geotechnical designs. Because laboratory and in-situ tests for determining the compression index (Cc) value are laborious, time consuming and costly, empirical formulas based on soil parameters are commonly used. Over the years a number of empirical formulas have been proposed to relate the compressibility to other soil parameters, such as the natural water content, the liquid limit, the plasticity index, the specific gravity. These empirical formulas provide good results for a specific test set, but cannot accurately or reliably predict the compression index from various test sets. The other disadvantage is that they tend to use a single parameter to estimate the compression index (Cc), even though Cc exhibits spatial characteristics depending on several soil parameters. This study presents the potential for Genetic Expression Programming (GEP) and the Adaptive Neuro-Fuzzy (ANFIS) computing paradigm to predict the compression index from soil parameters such as the natural water content, the liquid limit, the plastic index, the specific gravity and the void ratio. A total of 299 data sets collected from the literature were used to develop the models. The performance of the models was comprehensively evaluated using several statistical verification tools. The predicted results showed that the GEP and ANFIS models provided fairly promising approaches to the prediction of the compression index of soils and could provide a better performance than the empirical formulas.

Teaching visual arts : from the innocent eye to immersiveness and vice versa

Opis: In the mid-19th century, John Ruskin, apologist for the poetics of the innocent eye, advocated a break with academic Realism. A century later, Gombrich and Goodman criticized Ruskin's Romantic subjectivism from the position of High Modernism. Consequently, the "Enlightenment" approach in teaching visual arts became stronger. Recently, some teaching specialists have been trying to inaugurate the term, immersiveness, in place of visual arts language for the purpose of introducing contemporary art into the syllabus. This paper examines the relation between "the innocent eye myth" and topical approach of "immersiveness" within the framework of educational objectives and methods in contemporary teaching.
