3. The impact of weather conditions on alpha-acid content in hop (Humulus lupulus L.) cv. AuroraDouglas MacKinnon, Viljem Pavlovič, Barbara Čeh, Boštjan Naglič, Martin Pavlovič, 2020, izvirni znanstveni članek Opis: The influence of four main weather attributes on the content of alpha-acids of the hop cv. Aurora for the period 1994-2019 was studied. By analysing correlation coefficients, specific times of the year when the weather conditions affect the alpha-acid content with the goal of creating a forecasting model in Slovenia were identified. The most significant periods of weather that impacted the alpha-acid contents throughout the growing time of year are recognised as attributes of temperatures (T), rainfall (R) and sunshine (S) calculated from the 25th to 30th week (T2530, r = -0.78, P < 0.01; R2529, r = 0.72, P < 0.01 and S2529, r = -0.81, P < 0.01) and attributes of relative humidity (RH) from the 27th to 32nd week (RH2732, r = 0.82, P < 0.01). T2530 stands for the amount of active temperatures from June 18 to July 29. Likewise, R2530 matches to the precipitation (in mm or L/m2) during the same time period. 4. A Bayesian analysis of hop price fluctuationsDouglas MacKinnon, Martin Pavlovič, 2020, izvirni znanstveni članek Opis: This paper quantifies the correlation between U.S. season average prices for hops with U.S. hop stocks and U.S. hop hectarage. The Hop Equilibrium Ratio, a measure of the supply/demand relationship for U.S. hops, was introduced. Through the Bayesian inference method, the authors used these data to calculate the effect an incremental change to one metric had on the probability of directional changes of future U.S. season average prices (SAP). Between 2010 and 2020, the dominance of proprietary varieties created unprecedented cartel-like powers offering opportunities for supply- and price-management. Research results will enable more accurate forecasting and greater price stability in the hop industry. personalized therapy for Crohn’s disease (CD) patients. The motivation behind our study is to shift
the paradigm of anti-TNFα biomarker discovery toward specific immune cell sub-populations using
single-cell RNA sequencing and an innovative approach designed to uncover PBMCs gene expression
signals, which may be masked due to the treatment or ongoing inflammation; Methods: The singlecell
RNA sequencing was performed on PBMC samples from CD patients either naïve to biological
therapy, in remission while on adalimumab, or while on ustekinumab but previously non-responsive
to adalimumab. Sieves for stringent downstream gene selection consisted of gene ontology and
independent cohort genomic profiling. Replication and meta-analyses were performed using publicly
available raw RNA sequencing files of sorted immune cells and an association analysis summary.
Machine learning, Mendelian randomization, and oligogenic risk score methods were deployed to
validate DEGs highly relevant to anti-TNFα therapy response; Results: This study found PLCB1 in
CD4+ T cells and CRTAM in double-negative T cells, which met the stringent statistical thresholds
throughout the analyses. An additional assessment proved causal inference of both genes in response
to anti-TNFα therapy; Conclusions: This study, jointly with an innovative design, uncovered
novel candidate genes in the anti-TNFα response landscape of CD, potentially obscured by therapy

6. Modelling of dysregulated glucagon secretion in type 2 diabetes by considering mitochondrial alterations in pancreatic ▫$\alpha$▫-cellsVladimir Grubelnik, Rene Markovič, Saška Lipovšek Delakorda, Gerd Leitinger, Marko Gosak, Jurij Dolenšek, Ismael Valladolid-Acebes, Per-Olof Berggren, Andraž Stožer, Matjaž Perc, Marko Marhl, 2020, izvirni znanstveni članek Opis: Type 2 diabetes mellitus (T2DM) has been associated with insulin resistance and the failure of β-cells to produce and secrete enough insulin as the disease progresses. However, clinical treatments based solely on insulin secretion and action have had limited success. The focus is therefore shifting towards α-cells, in particular to the dysregulated secretion of glucagon. Our qualitative electron-microscopy-based observations gave an indication that mitochondria in α-cells are altered in Western-diet-induced T2DM. In particular, α-cells extracted from mouse pancreatic tissue showed a lower density of mitochondria, a less expressed matrix and a lower number of
cristae. These deformities in mitochondrial ultrastructure imply a decreased efficiency in mitochondrial ATP production, which prompted us to theoretically explore and clarify one of the most challenging problems associated with T2DM, namely the lack of glucagon secretion in hypoglycaemia and its oversecretion at high blood glucose concentrations. To this purpose, we constructed a novel computational model that links α-cell metabolism with their electrical activity and glucagon secretion. Our results show that defective mitochondrial metabolism in α-cells can
account for dysregulated glucagon secretion in T2DM, thus improving our understanding of T2DM pathophysiology and indicating possibilities for new clinical treatments.

7. Role of cAMP in double switch of glucagon secretionJan Zmazek, Vladimir Grubelnik, Rene Markovič, Marko Marhl, 2021, izvirni znanstveni članek Opis: Glucose metabolism plays a crucial role in modulating glucagon secretion in pancreatic alpha cells. However, the downstream effects of glucose metabolism and the activated signaling pathways influencing glucagon granule exocytosis are still obscure. We developed a computational alpha cell model, implementing metabolic pathways of glucose and free fatty acids (FFA) catabolism and an intrinsically activated cAMP signaling pathway. According to the model predictions, increased catabolic activity is able to suppress the cAMP signaling pathway, reducing exocytosis in a Ca2+ -dependent and Ca2+ independent manner. The effect is synergistic to the pathway involving ATPdependent closure of KATP channels and consequent reduction of Ca2+. We analyze the contribution
of each pathway to glucagon secretion and show that both play decisive roles, providing a kind of "secure double switch". The cAMP-driven signaling switch plays a dominant role, while the ATP-driven metabolic switch is less favored. The ratio is approximately 60:40, according to the most recent experimental evidence. Določanje molskih mas ▫$\alpha$▫-amilaze, ▫$\beta$▫-glukozidaze, proteaze in lipaze z metodo gelske kromatografije : magistrsko delo študijskega programa II. stopnjeJasmina Heric, 2019, magistrsko delo Opis: Gelska kromatografija je vrsta tekočinske kromatografije in je metoda za ločevanje molekul glede na molekulsko maso. Za gelsko filtracijo uporabljamo steklene kolone, napolnjene s stacionarno (trdno) fazo z enakomerno velikostjo por. Volumen oz. prostor med kroglicami gela (prazen volumen kolone) zapolnjuje mobilna (tekoča) faza.
Namen magistrske naloge je bil s pomočjo gelske kromatografije določiti molske mase encimov α-amilaze, β-glukozidaze, proteaze in lipaze. Najprej smo določili skupno koncentracijo proteinov v vzorcih ter določili elucijsi profil encimov. S pomočjo označevalcev znanih molekulskih mas (metilenskega modrila in dekstran modrega) smo izrisali umeritveno krivuljo za določanje molskih mas in nato določili molske mase encimov α-amilaze, β-glukozidaze, proteaze in lipaze. Ugotovili smo, da je eksperimentalno določena molska masa α-amilaze je 54,6 kDa, β-glukozidaze 112,2 kDa, proteaze 78,3 kDa in lipaze 26,6 kDa. Rezultate eksperimentalno določenih molskih mas smo primerjali s podatki o molskih masah najdenih v literaturi. Nato smo vse štiri encime združili v mešanico 4 encimov, katerim smo prav tako določili skupno koncentracijo proteinov s pomočjo Bradfordove metode in nato določili aktivnost posameznih encimov α-amilaze, β-glukozidaze, proteaze in lipaze v mešanici 4 encimov.

Learning and understanding new algorithms can be quite interesting, but also hard and complex, especially for students. Visualization can significantly help with the understanding of the dynamic behaviour of algorithms by visually displaying each step of the algorithm, its purpose and how it changes the data. Besides faster and more efficient learning, the better understanding can also lead to potential algorithm improvements in the future. The goal of this thesis is visualization of the alpha-beta tree search algorithm for determining the next optimal move in a two-player, zero-sum, complete information game. The algorithm is visualized using two games, Tic-Tac-Toe and Othello. The algorithm operation can also be demonstrated using a custom tree with parameters chosen by the user.
