Effect of Lactobacillus spp. on adhesion, invasion, and translocation of Campylobacter jejuni in chicken and pig small-intestinal epithelial cell lines

Opis: Background : Campylobacter spp. are a major cause of bacterial food-borne diarrhoeal disease. This mainly arises through contamination of meat products during processing. For infection, Campylobacter spp. must adhere to epithelial cells of the mucus layer, survive conditions of the gastrointestinal tract, and colonise the intestine of the host. Addition of probiotic bacteria might promote competitive a dhesion to epithelial cells, consequently reducing Campylobacter jejuni colonisation. Effect of Lactobacillus spp. (PCS20, PCS22, PCS25, LGG, PCK9) on C. jejuni adhesion, invasion and translocation in pig (PSI cl.1) and chicken (B1OXI) small-intestine cell lines, as well as pig enterocytes (CLAB) was investigated. Results : Overall, in competitive adhesion assays with PSI cl.1 and CLAB cell monolayers, the addition of Lactobacillus spp. reduced C. jejuni adherence to the cell surface, and negatively affected the C. jejuni invasion. Interestingly, Lactobacillus spp. significantly impaired C. jejuni adhesion in three-dimensional functional PSI cl.1 and B1OXI cell models. Also, C. jejuni did not translocate across PSI cl.1 and B1OXI cell monolayers when co-incubated with probiotics. Among selected probiotics, Lactobacillus rhamnosus LGG was the strain that reduced adhesion efficacy of C. jejuni most significantly under co-culture conditions. Conclusion : The addition of Lactobacillus spp. to feed additives in livestock nutrition might be an effective novel strategy that targets Campylobacter adhesion to epithelial cells, and thus prevents colonisation, reduces the transmission, and finally lowers the incidence of human campylobacteriosis.

Exploring chitosan-plant extract bilayer coatings: Advancements in active food packaging via polypropylene modification

Opis: UV-ozone activated polypropylene (PP) food films were subjected to a novel bilayer coating process involving primary or quaternary chitosan (CH/QCH) as the first layer and natural extracts from juniper needles (Juniperus oxycedrus; JUN) or blackberry leaves (Rubus fruticosus; BBL) as the second layer. This innovative approach aims to redefine active packaging (AP) development. Through a detailed analysis by surface characterization and bioactivity assessments (i.e., antioxidant and antimicrobial functionalities), we evaluated different coating combinations. Furthermore, we investigated the stability and barrier characteristics inherent in these coatings. The confirmed deposition, coupled with a comprehensive characterization of their composition and morphology, underscored the efficacy of the coatings. Our investigation included wettability assessment via contact angle (CA) measurements, X-ray photoelectron spectroscopy (XPS), and attenuated total reflectance Fourier-transform infrared spectroscopy (ATR-FTIR), which revealed substantial enhancements in surface concentrations of elements and functional groups of CH, QCH, JUN, and BBL. Scanning electron microscopy (SEM) unveiled the coatings' heterogeneity, while time-of-flight secondary ion mass spectrometry (ToF-SIMS) and CA profiling showed moderately compact bilayers on PP, providing active species on the hydrophilic surface, respectively. The coatings significantly reduced the oxygen permeability. Additionally, single-layer depositions of CH and QCH remained below the overall migration limit (OML). Remarkably, the coatings exhibited robust antioxidative properties due to plant extracts and exceptional antimicrobial activity against S. aureus, attributed to QCH. These findings underscore the pivotal role of film surface properties in governing bioactive characteristics and offer a promising pathway for enhancing food packaging functionality.

Razvoj novih funkcionalnih plastičnih filmov za različne produkte : magistrsko delo

Opis: Plastični materiali imajo velik pomen v pakirnem segmentu za pakiranje farmacevtskih, kozmetičnih in prehrambnih produktov, kjer želimo podaljšati rok trajanja živil. Vedno večje zavedanje podjetij in potrošnikov o negativnem vplivu petrokemijske plastike na okolje in povečano tveganje za zdravje, ki ga predstavljajo sintetični aditivi v plastični embalaži, so privedli do razvoja biorazgradljive aktivne embalaže. V magistrskem delu smo želeli razviti antioksidativne, protimikrobne in biorazgradljive funkcionalne plastične filme. Folije PLA smo funkcionalizirali s premazom iz formulacije ekstrakta brina in hitozana, ki smo ga na površino folije nanesli s petimi različnimi metodami, in sicer z metodo pršenja, valjčkanja, omakanja, impregniranja in tehnologijo superkritičnih fluidov. Učinkovitost uporabljenih snovi in metod smo preučili z analiznimi metodami: FTIR, goniometrija, pregradne lastnosti – prepustnost kisika, antioksidativnost, XPS, SEM, transparentnost, protimikrobna aktivnost ter LCA in LCC analiza. Z analiznimi metodami smo potrdili odlično antioksidativno delovanje ekstrakta brina in protimikrobno delovanje hitozana. Izkazalo se je, da je za praktično uporabo z vidika okoljske in cenovne ustreznosti najprimernejši funkcionalni plastični film, kjer smo premaz nanesli z metodo omakanja. Dosegli smo odlično protimikrobno delovanje predvsem na bakterijo Staphylococcus aureus, izvrstne pregradne in antioksidativne lastnosti, potrjene s FTIR, XPS in SEM analizo, ter zelo dobre protikondenzacijske lastnosti.

Seasonal changes in chemical profile and antioxidant activity of Padina pavonica extracts and their application in the development of bioactive chitosan/PLA bilayer film

Opis: Seaweeds are a potentially sustainable source of natural antioxidants that can be used in the food industry and possibly for the development of new sustainable packaging materials with the ability to extend the shelf-life of foods and reduce oxidation. With this in mind, the seasonal variations in the chemical composition and antioxidant activity of brown seaweed (Padina pavonica) extracts were investigated. The highest total phenolic content (TPC) and antioxidant activity (measured by ferric reducing/antioxidant power (FRAP), 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging, and oxygen radical absorbance capacity (ORAC)) were found for P. pavonica June extract. The TPC of 26.69 ± 1.86 mg gallic acid equivalent/g, FRAP of 352.82 ± 15.41 µmole Trolox equivalent (TE)/L, DPPH of 52.51 ± 2.81% inhibition, and ORAC of 76.45 ± 1.47 µmole TE/L were detected. Therefore, this extract was chosen for the development of bioactive PLA bilayer film, along with chitosan. Primary or quaternary chitosan was used as the first layer on polylactic acid (PLA) films. A suspension of chitosan particles with entrapped P. pavonica extract was used as the second layer. X-ray photoelectron spectroscopy confirmed the presence of layers on the material surface. The highest recorded antioxidant activity of the newly developed films was 63.82% inhibition. The developed functional films exhibited antifogging and antioxidant properties, showing the potential for application in the food industry.
