1. Microbiota in vitro modulated with polyphenols shows decreased colonization resistance against Clostridioides difficile but can neutralize cytotoxicityAleksander Mahnič, Jennifer Auchtung, Nataša Poklar Ulrih, Robert A. Britton, Maja Rupnik, 2020, izvirni znanstveni članek Opis: While the knowledge on gut microbiota - C. difficile interactions has improved over the years, the understanding of the underlying mechanisms providing colonization resistance as well as preventative measures against the infection remain incomplete. In this study the antibiotic clindamycin and polyphenol extracts from pomegranate and blueberries were used individually and in combination to modulate fecal microbial communities in minibioreactor arrays (MBRA). Modulated communities were inoculated with C. difficile (ribotype 027). Subsequent 7-day periodical monitoring included evaluation of C. difficile growth and activity of toxins TcdA and TcdB as well as analysis of MBRA bacterial community structure (V3V4 16%S metagenomics). Polyphenols affected multiple commensal bacterial groups and showed different synergistic and antagonistic effects in combination with clindamycin. Exposure to either clindamycin or polyphenols led to the loss of colonization resistance against C. difficile. The successful growth of C. difficile was most significantly correlated with the decrease in Collinsella and Lachnospiraceae. Additionally, we demonstrated that Clostridium sporogenes decreased the activity of both C. difficile toxins TcdA and TcdB. The feature was shown to be common among distinct C. sporogenes strains and could potentially be applicable as a non-antibiotic agent for the alleviation of C. difficile infection. 3. Two-layer functional coatings of chitosan particles with embedded catechin and pomegranate extracts for potential active packagingSanja Potrč, Tjaša Kraševac Glaser, Alenka Vesel, Nataša Poklar Ulrih, Lidija Fras Zemljič, 2020, izvirni znanstveni članek Opis: Two-layer functional coatings for polyethylene (PE) and polypropylene (PP) films were developed for the active packaging concept. Prior to coating, the polymer films were activated by O$_2$ and NH$_3$ plasma to increase their surface free energy and to improve the binding capacity and stability of the coatings. The first layer was prepared from a macromolecular chitosan solution, while the second (upper) layer contained chitosan particles with embedded catechin or pomegranate extract. Functionalized films were analyzed physico-chemically to elemental composition using ATR-FTIR spectroscopy and XPS. Further, oxygen permeability and wettability (Contact Angle) were examined. The antimicrobial properties were analyzed by the standard ISO 22196 method, while the antioxidative properties were determined with an ABTS assay. Functionalized films show excellent antioxidative and antimicrobial efficacy. A huge decrease in oxygen permeability was achieved in addition. Moreover, a desorption experiment was also performed, confirming that the migration profile of a compound from the surfaces was in accordance with the required overall migration limit. All these properties indicate the great potential of the developed active films/foils for end-uses in food packaging. 4. Razvoj kompozitnega materiala s kanabidiolom in celulozo : magistrsko deloKaja Vrezner, 2020, magistrsko delo Opis: Veliko žensk se med menstruacijo bori z menstrualnimi krči. Da bi pripomogli k rešitvi tega problema, želimo izdelati tampon, s kanabidiolom (CBD), kateri ima zelo velik potencial v zdravstvu. Poleg tega gre za naravno in biorazgradljivo spojino. Namreč veliko sintetičnih zdravil se nepravilno odlaga, kar ima na okolje negativen vpliv. Z nano-kapsulacijo smo poskušali ujeti CBD v liposome, saj ti izboljšajo absorpcijo kanabidola. Učinkovitost kapsulacije smo preverjali z FTIR spektroskopijo, meritvami velikosti delcev, elektronsko mikroskopijo in GC-MS analizno metodo. Rezultati so pokazali, da se je CBD ujel v liposome. Tako pripravljeno formulacijo smo nanesli neposredno na celulozna vlakna. Analiza je pokazala, da je funkcionaliziran celulozni vzorec antioksidativen, protimikroben in sposoben desorpcije CBD.
