Antibacterial effect of topically administered tranexamic acid in large joint arthroplasty

Opis: Tranexamic acid (TXA) is a synthetic derivative of the amino acid lysine and is commonly used as an antifibrinolytic agent in large joint surgery, such as knee arthroplasty, to minimize blood loss and its associated complications. Some studies have suggested that perioperative intravenous TXA application lowers the incidence of postoperative infections after hip and knee arthroplasty. While its effects on bacteria are not its primary purpose, more research is needed further to elucidate the extent of TXA’s impact on bacterial inhibition and its clinical relevance. In order to determine possible inhibitory effects, 22 clinically relevant bacterial strains were exposed to different TXA concentrations. Two different assays were performed: a microdilution assay with traditional endpoint measurements and a kinetic growth assay for a time-dependent susceptibility assessment. With the microdilution technique, a growth inhibitory effect became evident at higher TXA concentrations (50 and 100 mg/mL) on eight tested bacterial strains, of which four demonstrated absence of growth at 100 mg/mL, suggesting minimal inhibitory concentration. However, the kinetic growth assay revealed more specific TXA-induced dynamic changes in the bacterial growth of all tested bacterial strains. This study showed that TXA affects the growth dynamics of bacteria by altering their lag phase duration, exponential growth rate, and population density and may contribute to the prevention of implant-associated infections.

The immobilization of β-galactosidase on glass fiber rolls

Opis: The usability of glass fibers as immobilization support with a porous open structure was investigated. We developed a method to immobilize the enzyme β-galactosidase on special glass fiber rolls. The new method is simple, non-expensive and industrially applicable. Glutaraldehyde was used as a non-specific cross-linking agent for the covalent binding of β-galactosidase on modified glass fibers. The efficiency of immobilization was tested with the known hydrolysis of lactose. All experiments were performed in a continuous laboratory reactor. The influence of the reaction temperature (20, 25 and 30 °C), the substrate flow rate (1, 2 and 3 mL/min) and the pH of the reaction medium (6, 7 and 8) on the conversion was investigated. The reaction efficiency was monitored by measuring the glucose concentration with a spectrophotometer. High immobilization efficiency, enzyme activity and stability were obtained. The optimal reaction temperature, substrate flow rate and pH were found. The activity and stability of the enzyme entrapped on the glass fiber rolls remained almost unchanged during reuse, which is promising for potential industrial applications.

Policijsko delo v skupnosti in partnersko zagotavljanje varnosti v lokalnih skupnostih

Opis: Monografija predstavlja policijsko delo v skupnosti v kontekstu zagotavljanja varnosti v lokalnih skupnostih v Sloveniji. Uvodoma so predstavljene teoretske perspektive policijskega dela v skupnosti. Sledi sistematični pregled evropskih raziskav o policijskem delu v skupnosti v zadnjih dveh desetletjih. Avtorji obravnavajo povezanost med legitimnostjo in policijskim delom v skupnosti v perspektivi javnega mnenja o policiji ter samozaznave legitimnosti policistov in njihovih pogledov na policijsko organizacijo. Posebno pozornost avtorji namenjajo opravljanju policijskega dela v skupnosti v urbanih in ruralnih okoljih, ker se narava kriminalitete in izvajanje policijske dejavnosti pomembno razlikuje med urbanimi in ruralnimi okolji. Sledi predstavitev usmerjenih skupinskih intervjujev s policisti in individualnih polstrukturiranih intervjujev s predstavniki lokalnih skupnosti. V naslednjem poglavju avtorji predstavijo analizo SWOT o policijskem delu v skupnosti. Avtorji pisanje zaključijo z refleksijo razvoja policijskega dela v Sloveniji v zadnjih treh desetletjih.

Don't leave your heart at work : profiles of work-life interference and cardiometabolic risk

Opis: The present study adopts an exploratory, person-oriented approach to investigate possible patterns of work-life interference. We examine work–life interference from a cognitive (i.e., thinking about work), behavioral (i.e., engaging in work-related behavior), and performance perspective (i.e., reduced functionality in private life, or work–life conflict) in order to identify profiles of employees that could potentially remain uncovered with variable-oriented research. Furthermore, as work–life interference relates to wellbeing and health, we were interested in exploring possible differences between profiles in emotional exhaustion, cardiometabolic risk, and health-related behavior. Self-report data on work–life interference and well-being, as well as objective health data, were collected from a heterogeneous sample of 289 employees. Four profiles with different patterns of work–life interference were identified. Out of the four profiles, two profiles reported moderate and high work–life interference (the Moderate Interference and High Interference profiles). The other two profiles revealed distinct combinations of moderate and low performance and behavioral interference (the Low Performance Interference and Low Behavioral Interference profiles). The High Interference and Low Behavioral Interference profiles were identified as risk groups in terms of cardiometabolic health, while the Low Performance Interference and Moderate Interference profiles showed low to no risk. Regarding work-related well-being, the High Interference profile showed the highest risk of emotional exhaustion.

Distrustful, dissatisfied, and conspiratorial : a latent profile analysis of COVID-19 vaccination rejection

Opis: Trust in science and scientists, satisfaction with the national government, and endorsement of conspiracy theories are important factors in the decision to be vaccinated. In this study, we investigated whether there are different profiles of individuals depending on the above factors and whether they differ in their sociodemographic composition. We used data from Round 10 of the European Social Survey for Slovenia, employing a nationally representative sample of 1252 participants. Based on latent profile analysis, three distinct profiles emerged: Profile 1 expressed moderate trust in science, satisfaction with government, and high endorsement of conspiracies; Profile 2 expressed low trust and satisfaction and moderate endorsement of conspiracies; Profile 3 expressed high trust and satisfaction and low beliefs in conspiracy theories. In addition, Profile 3 expressed the strongest support for vaccination and Profile 2 the lowest. Our results suggest that distrust, dissatisfaction, and the presence of conspiracy theories are the “perfect storm” for vaccination rejection. In contrast, despite conspiracy theories, a certain level of trust and satisfaction may reduce vaccination rejection.

Učinkovitost zdravljenja sindroma policističnih jajčnikov z metforminom

Opis: Metformin is an oral antidiabetic drug with potentially beneficial effects in patients with polycystic ovary syndrome (PCOS). Different pharmacological responses to metformin have been reported in PCOS patients, which may be attribut able to various clinical, biochemical, genetic and other factors. Here, we have outlined a short review of the results from current clinical studies on metformin efficacy in PCOS patients. Additionally, preliminary results from a retrospective pharmacogenetic study by our group on a sample of Slovene PCOS patients are presented. Pharmacogenetics is an emerging science that should provide options for individualized medicine, the future of healthcare.

Ideological differences in COVID-19 vaccine intention : the effects of trust in the healthcare system, in complementary and alternative medicine, and perceived threat from the disease

Opis: Introduction: Politically left-leaning individuals are more likely to get vaccinated against COVID-19, although little is known about the mechanisms underlying the ideological differences in vaccination intentions. Understanding the extent to which trust in the healthcare system, in complementary and alternative medicine, and the perceived threat from the disease contribute to these disparities is crucial, as it could inform targeted interventions to address vaccine hesitancy across the political spectrum. Methods: The present cross-sectional study conducted among adults living in Slovenia (n  =  858) examined the mediating role of trust in the healthcare system, trust in complementary and alternative medicine (CAM), and the perceived threat from the virus on COVID-19 vaccination intention. Results: We found that leftist ideology and trust in the healthcare system positively predicted vaccination intention, whereas CAM negatively predicted this intention. In addition, left-leaning individuals expressed greater trust in the healthcare system and lower trust in CAM, resulting in higher levels of COVID-19 vaccination intention. The serial mediation model confirmed that trust in CAM was a negative predictor, while trust in the healthcare system positively predicted perceived threat. Discussion: When dealing with vaccine hesitancy among right-oriented individuals, strategies should focus on enhancing trust in the healthcare system and critically evaluating the reliance on CAM.

Optimiranje postopkov formulacije farmacevtskih učinkovin v porozne polimerne nosilce

Opis: Namen magistrske naloge je bil optimizirati formulacijo polimernih nosilcev kot sistemov za dostavo farmacevtskih učinkovin. Cilj je bil izboljšati topnost izbranih farmacevtskih učinkovin ter doseči podaljšano sproščanje v simuliranih telesnih tekočinah. Polimerni nosilci so obetavni na področju razvoja sistemov s formuliranimi zdravili, saj lahko z ustrezno prilagoditvijo njihovih fizikalno-kemijskih lastnosti dosežemo izboljšano formuliranje in sproščanje farmacevtskih učinkovin. V sklopu laboratorijskega dela smo sintetizirali plastovite polimerne nosilce, v katere smo uspešno formulirali dve različni farmacevtski učinkovini z različno polarnostjo. Izbrana polimera sta bila hitozan in alginat. S procesom superkritičnega penjenja smo izdelali tudi porozne polimerne nosilce, kjer smo kot začetni polimer uporabili polikaprolakton. Kot farmacevtski učinkovini smo izbrali ketoprofen in klindamicin hidroklorid. Izbrali smo ju glede na njuno topnost v telesnih tekočinah. Ketoprofen se uporablja za zdravljenje akutne bolečine in je slabo topen, medtem ko je klindamicin hidroklorid bolj topen v telesnih tekočinah in se uporablja kot antibiotik. Karakterizacijo formuliranih polimernih nosilcev z vsebnostjo farmacevtskih učinkovin smo izvedli z vrstično elektronsko mikroskopijo in tehniko oslabljenega totalnega odboja infrardeče spektroskopije s Fourierjevo transformacijo. Z in vitro testiranjem sproščanja smo prikazali spreminjanje vsebnosti formuliranih farmacevtskih učinkovin v polimernih nosilcih glede na čas sproščanja, pridobljene vzorce smo analizirali s tekočinsko kromatografijo visoke ločljivosti in z UV-Vis spektrofotometrijo. Rezultati so nedvoumno pokazali, da je formulacija farmacevtskih učinkovin v polimerne nosilce omogočila podaljšano sproščanje v telesnih tekočinah. Ugotovitve kažejo, da so porozni polimerni nosilci bolj primerni za sisteme dostave zdravil s podaljšanim sproščanjem v primerjavi s plastovitimi polimernimi nosilci.
