Human beta cell functional adaptation and dysfunction in insulin resistance and its reversibility

Abstract: Background: Beta cells play a key role in the pathophysiology of diabetes since their functional adaptation is able to maintain euglycemia in the face of insulin resistance, and beta cell decompensation or dysfunction is a necessary condition for full-blown type 2 diabetes (T2D). The mechanisms behind compensation and decompensation are incompletely understood, especially for human beta cells, and even less is known about influences of chronic kidney disease (CKD) or immunosupressive therapy after transplantation on these processes and the development of posttransplant diabetes. Summary: During compensation, beta cell sensitivity to glucose becomes left-shifted, i.e., their sensitivity to stimulation increases, and this is accompanied by enhanced signals along the stimulus-secretion coupling cascade from membrane depolarization to intracellular calcium and the most distal insulin secretion dynamics. There is currently no clear evidence regarding changes in intercellular coupling during this stage of disease progression. During decompensation, intracellular stimulus-secretion coupling remains enhanced to some extent at low or basal glucose concentrations but seems to become unable to generate effective signals to stimulate insulin secretion at high or otherwise stimulatory glucose concentrations. Additionally, intercellular coupling becomes disrupted, lowering the number of cells that contribute to secretion. During progression of CKD, beta cells also seem to drift from a compensatory left-shift to failure, and immunosupressants can further impair beta cell function following kidney transplantation. Key Messages: Beta cell stimulus-secretion coupling is enhanced in compensated insulin resistance. With worsening insulin resistance, both intra- and intercellular coupling become disrupted. CKD can progressively disrupt beta cell function, but further studies are needed, especially regarding changes in intercellular coupling.

Destination climate adaptation

Abstract: A key element in the product mix of destinations is climate. Climate represents a critical part of a destination's economic and resource base such that changes in climate will trigger human responses in terms of demand and the type of activities that the climate will support. This threatens the competitiveness, sustainability, and economic viability of destinations. This research note focuses on destination adaptation to climate change that is anticipatory not reactive, based on projecting future climate scenarios for a destination and then assessing the tourism products that the future climate will support. It outlines an original data-driven approach to adaptation that is generalizable to other destinations. The research note describes an exploratory research collaboration in Croatia between tourism and climate scientists that allows, first, the modeling of a destination's projected climate conditions and, second, the products and activities that can be supported by these climate scenarios using climate indices for tourism.

Supply chain management mitigation to climate change in three selected industrial sectors

Abstract: As the world economy is growing and globalisation is on the rise, climate change and its effects on the planet Earth are also increasing. But climate changes do not disturb only nature; they also affect businesses and their supply chains. Many companies have recently recognised the positive impacts of green and circular supply chain management practices. Supply chains that deliver products from around the world are increasingly exposed to the impacts of a changing climate and scarce resources. In this research, it was determined how climate change impacts supply chains in the food, automobile, and energy industry sectors. Furthermore, we wanted to find out if and how the management of supply chains is altered to adapt to the business environment which is more and more affected by environmental changes.

Impact of transport trends on sustainability in the Western Balkans : a future-oriented business sector perspective

Abstract: Abstract This study examines the impact of emerging transport trends on the Western Balkans and their potential to improve the sustainability and efficiency of the transport sector in the region. In the context of global efforts to reduce environmental impacts and improve transport safety, the study aims to better understand the perspective of the business sector in order to align strategic planning for the introduction of new transport technologies and practices with the expectations of the business sector. Using a questionnaire-based survey on the Mentimeter platform, the study gathers insights from 49 transport and logistics experts. The analysis uses violin plots and heat maps to visualise the expected impact of the identified trends. The results show a consensus among the experts that the emerging trends are likely to lead to a reduction in accidents, congestion, and emissions, while at the same time increasing operating costs and investment in infrastructure. In particular, trends such as alternative fuels, electrification, and smart city initiatives are highlighted as important drivers for reducing emissions. The study concludes that while these trends are likely to lead to the achievement of environmental and safety goals, they also lead to economic challenges as operating costs increase and significant infrastructure investment is required. The study emphasises the need for strategic investment, policy adjustments, and capacity building to lead the Western Balkans towards a sustainable transport future.

Implementation of climate change effects on slope stability analysis

Abstract: The objective of this study is to determine the impacts of expected climate change on slope stability. For this purpose, the case study of a slope instability, that was triggered in 2021 was selected. The stability analysis was performed considering the theory of rainfall infiltration and using Geo-Studio’s SEEP/W module for the surface infiltration model of the slope. A parametric stability analysis of the slope was conducted to determine the importance of climate change on slope stability. Conditions for changes in volumetric water content, water permeability, porewater pressure, and groundwater flow are important. When soil permeability is low, the factor of safety decreases during rainfall events and on the days following, while when permeability is higher, safety increases after rainfall events. The effect of lower cohesion is nearly linear, with the factor of safety decreasing by 0.1 for every 1 kPa less cohesion. The increase in net infiltration of water may be the most critical factor for slope instability. The results of the analysis indicate that timely reduction of water net infiltration through planting and proper surface water runoff from the upper road and slope would be a relatively simple and inexpensive measure compared to the cost of remediating the landslide, considering expected climate change. Therefore, it is advisable to analyze all slopes with respect to the expected climate change, taking into account the potential impacts of climate change.

Operatic adaptation of Shakespeare`s Romeo and Juliet

Abstract: This Master’s Thesis analyses Charles Gounod’s operatic adaptation of William Shakespeare’s Romeo and Juliet. The theoretical part introduces the theory of adaptation with a particular focus on operatic adaptations; it then moves on to three modes of adaptations, and discusses the correlation between adaptation and plagiarism. In the second chapter, the thesis identifies and interprets the sources for Shakespeare’s and Gounod’s works, introduces the librettists Jules Barbier and Michael Carré and discusses the background of the period of the operatic production. The main purpose of this thesis is to identify and study stylistic correlation between Shakespeare’s Romeo and Juliet and the libretto of Roméo et Juliette. The thesis further establishes whether the librettists make certain themes from the play more or less prominent. For this purpose, three passages were selected for the contrastive microstructural analysis of the play and its operatic adaptation in the final chapter of this thesis. The intent is to establish whether librettists managed to preserve Shakespeare’s most salient stylistic elements as well as other textual characteristics that have an impact on the interpretative potential of the adaptation.

Abolition of exequatur in Brussels Ia regulation : new challenges for the National Judge (Croatia)

Abstract: This article will analyse and evaluate if and to what extent enforcement of judgments according to the Brussels Ia Regulation may be challenging for Croatian judges. It seeks to answer the questions a) which novelties in terms of recognition and enforcement of foreign judgments are introduced through the system of cross-border enforcement of judgments under Brussels Ia and b) with regard to the lack of implementation provisions in Croatian legal system, how will a new “adaptation device” according to Art. 54 of the Brussels operate. In particular, how and by whom will the adaptation of measure or order be carried out and how will it be challenged pursuant to Art. 54 (2) of the Brussels Ia Regulation. In a search for possible answers, approaches as well as solutions adopted in the legal systems of some Member States will also be taken into account. Considerations which can be attributed to the lack of provisions regarding implementation of the Brussels Ia Regulation in Croatian legal system will be highlighted.

Abstract: The present scientific monograph, entitled “Environmental management & audit“, is the result of three years’ work on an international project entitled “Environmental management in Russian companies – retraining courses for the sensibilization for and integration of Eco-Audit programs in corporate decision-making (RECOAUD)”. Within its more than 600 pages, the monograph features interesting texts written by 31 authors from the European Union and the Russian Federation, edited by dr. Borut Jereb, Darja Kukovič and dr. Daria Meyr. The monograph “Environmental management & audit“ is composed of four books: “Scarcity and Introduction of Environmental Management”, “Management Systems”, “Controlling and Stakeholders”, and “Environmental Assessment” (Featured Articles). These four topics reflect the complexity, heterogeneity and multidisciplinary of the project Tempus RECOAUD. The reader of the monograph gets a comprehensive overview of theoretical perspectives of environmental management and audit in the chosen areas. Furthermore, the monograph also highlights the results of research in the field of environmental management and audit as well as trends and challenges in the development of this field. Providing insight into theoretical and research findings, the monograph will prove useful to both practitioners and researchers in the field of environmental management and audit; it can also be used for study purposes.

An Analysis of the Dramatization of 19th Century Medical Science in the Television Show The Knick

Abstract: I researched how accurate the dramatization of medical science is in the television series The Knick. The thesis is separated into two parts. In the first part, I analyze the accuracy of the depiction of real persons in the series. The main character John Thackery is an adaptation of the real-life surgeon William Halsted. The character of Dr. Cotton is a dramatization of the real psychiatrist named Henry Cotton. The series also adapts the story of Mary Mallon, who was a healthy carrier of typhoid fever and was responsible for spreading the disease in households where she worked as a cook. In the second part of my thesis, I analyze the accuracy of the medical science depicted in the series and explain how the producers used visual effects to achieve historical accuracy.
