Discovery of novel biomarkers with extended non-coding RNA interactor networks from genetic and protein biomarkers

Abstract: Curated online interaction databases and gene ontology tools have streamlined the analysis of highly complex gene/protein networks. However, understanding of disease pathogenesis has gradually shifted from a protein-based core to complex interactive networks where non-coding RNA (ncRNA) is thought to play an essential role. As current gene ontology is based predominantly on protein-level information, there is a growing need to analyze networks with ncRNA. In this study, we propose a gene ontology workflow integrating ncRNA using the NPInter V5.0 database. To validate the proposed workflow, we analyzed our previously published curated biomarker datasets for hidden disease susceptibility processes and pharmacogenomics. Our results show a novel involvement of melanogenesis in psoriasis response to biological drugs in general. Hyperpigmentation has been previously observed in psoriasis following treatment with currently indicated biological drugs, thus calling attention to melanogenesis research as a response biomarker in psoriasis. Moreover, our proposed workflow highlights the need to critically evaluate computed ncRNA interactions within databases and a demand for gene ontology analysis of large miRNA blocks.

Meta-analytic comparison of global RNA transcriptomes of acute and chronic myeloid leukemia cells reveals novel gene candidates governing myeloid malignancies

Abstract: Despite advances in the understanding of genetic risk factors and molecular mechanisms underlying acute myeloid leukemia (AML) and chronic myeloid leukemia (CML), clinical outcomes of current therapies in terms of disease relapse and mortality rate pose a great economic and social burden. To overcome this, the identification of new molecular prognostic biomarkers and pharmacological targets is crucial. Recent studies have suggested that AML and CML may share common pathogenic mechanisms and cellular substrates. To this end, in the present study, global transcriptome profiles of AML and CML at the molecular and cellular level were directly compared using a combination of meta-analysis and modern statistics, and novel candidate genes and specific biological processes associated with the pathogenesis of AML and CML were characterized. Our study significantly improves our current understanding of myeloid leukemia and will help develop new therapeutic targets and biomarkers for disease progression, management and treatment response.

Logistični procesi pri nabavi kemijskih materialov v podjetju Hisense Gorenje Europe

Abstract: Diplomsko delo smo razdelili na dva dela: teoretični in praktični. V teoretičnem delu smo opredelili, kaj je to logistika, opisali logistične podsisteme in elemente logistike s poudarkom na nabavni logistiki. V praktične delu pa se bomo omejili na logistične procese pri nabavi kemijskih elementov v podjetju Hisense Gorenje Europe. Metode, ki smo jih pri raziskovalnem delu uporabili bodo poslovne, saj bomo v diplomski nalogi proučevali poslovno (nabavno) funkcijo podjetja. Uporabljene metode se lahko opredeli tudi kot dinamične, saj se poslovne funkcije spreminjajo in prilagajajo okoliščinam poslovanja. K raziskovanju smo pristopili deskriptivno, saj bomo s pomočjo strokovne literature in spletnih virov opredelili logistiko, opisali njene podsisteme in elemente. Podatke za pripravo diplomskega dela smo pridobili s pomočjo intervjuvanja mentorja, ki nam ga je dodelilo podjetje. Hipoteza iz katere smo izhajali je bila, da dobro organizirana nabavna logistika pozitivno vpliva na delovanje podjetja Hisense Gorenje Europe.

Identification of novel loci involved in adalimumab response in Crohn’s disease patients using integration of genome profiling and isoform-level immune-cell deconvoluted transcriptome profiling of colon tissue

Abstract: Crohn’s disease is a consequence of dysregulated inflammatory response to the host’s microbiota. Although anti-TNF treatment improves the quality of the patient’s life, a large proportion of patients lose response to the treatment. The past decade of research has led to a continuum of studies showcasing the heterogeneity of anti-TNF response; thus, the aim of the present study was to dissect transcriptome-wide findings to transcript isoform specific levels and combine the analyses with refined information of immune cell landscapes in colon tissue, and subsequently select promising candidates using gene ontology and genomic integration. We enrolled Slovenian Crohn’s disease patients who were naïve with respect to adalimumab treatment. We performed colon tissue RNA sequencing and peripheral blood mononuclear cell DNA genotyping with a subsequent contemporary integrative approach to combine immune cell deconvoluted isoform transcript specific transcriptome analysis, gene ontology layering and genomic data. We identified nine genes (MACF1, CTSE, HDLBP, HSPA9, HLA-DMB, TAP2, LGMN, ANAPC11, ACP5) with 15 transcripts and 16 variants involved in the adalimumab response. Our study identified loci, some of which were previously shown to contribute to inflammatory bowel disease susceptibility, as novel loci involved in adalimumab response in Crohn’s disease patients.

Brain dynamics underlying preserved cycling ability in patients with Parkinson’s disease and freezing of gait

Abstract: Parkinson’s disease (PD) is generally associated with abnormally increased beta band oscillations in the cortico-basal ganglia loop during walking. PD patients with freezing of gait (FOG) exhibit a more distinct, prolonged narrow band of beta oscillations that are locked to the initiation of movement at ∼18 Hz. Upon initiation of cycling movements, this oscillation has been reported to be weaker and rather brief in duration. Due to the suppression of the overall beta band power during cycling and its continuous nature of the movement, cycling is considered to be less demanding for cortical networks compared to walking, including reduced need for sensorimotor processing, and thus unimpaired continuous cycling motion. Furthermore, cycling has been considered one of the most efficient non-pharmacological therapies with an influence on the subthalamic nucleus (STN) beta rhythms implicative of the deep brain stimulation effects. In the current review, we provide an overview of the currently available studies and discuss the underlying mechanism of preserved cycling ability in relation to the FOG in PD patients. The mechanisms are presented in detail using a graphical scheme comparing cortical oscillations during walking and cycling in PD.

Review of potential drug-eluting contact lens technologies

Abstract: The field of ophthalmology is expanding exponentially, both in terms of diagnostic and therapeutic capabilities, as well as the worldwide increasing incidence of eye-related diseases. Due to an ageing population and climate change, the number of ophthalmic patients will continue to increase, overwhelming healthcare systems and likely leading to under-treatment of chronic eye diseases. Since drops are the mainstay of therapy, clinicians have long emphasised the unmet need for ocular drug delivery. Alternative methods, i.e., with better compliance, stability and longevity of drug delivery, would be preferred. Several approaches and materials are being studied and used to overcome these drawbacks. We believe that drug-loaded contact lenses are among the most promising and are a real step toward dropless ocular therapy, potentially leading to a transformation in clinical ophthalmic practice. In this review, we outline the current role of contact lenses in ocular drug delivery, focusing on materials, drug binding and preparation, concluding with a look at future developments.

Effects of cardiac rehabilitation and diet counselling on adherence to the mediterranean lifestyle in patients after myocardial infarction

Abstract: Adherence to the Mediterranean lifestyle—as captured by the Medlife Index Questionnaire (i.e., encompassing a Mediterranean diet as well as other aspects of healthy living, such as food preparation, physical activity, and socializing)—has been associated with reduced cardiovascular events in healthy individuals. In the present study, we sought to determine the adherence to, and the effect of comprehensive cardiac rehabilitation on, Mediterranean lifestyle adherence in patients after myocardial infarction. We included 121 patients (mean age, 55 years; women, 37%) undergoing comprehensive cardiac rehabilitation—i.e., exercise training 3 times per week for 12 weeks plus dedicated workshops promoting the Mediterranean lifestyle. Before and after cardiac rehabilitation, patients completed the Medlife Index Questionnaire. High baseline adherence was associated with favourable glucose (5.39 vs. 6.1 mmol/L; p < 0.001), triglycerides (1.1 vs. 1.5 mmol/L; p = 0.002), and HDL cholesterol levels (1.32 vs. 1.12 mmol/L; p = 0.032). More importantly, the Medlife Score significantly improved following comprehensive cardiac rehabilitation in patients with low baseline adherence (from 13.8 to 16.7 points; p < 0.001), but not in patients with high baseline adherence (from 19.4 to 18.8 points; p = 0.205). Our findings suggested that Mediterranean lifestyle promotion during cardiac rehabilitation improved adherence to the Mediterranean lifestyle, especially in low-adherence patients.

Ključni kriteriji, ki vplivajo na odločitve kupcev za spletno nakupovanje : diplomsko delo

Abstract: Izbira najbolj primernega izdelka ali storitve prek spleta je lahko izčrpen proces, saj je široka raznolikost izdelkov na spletnih straneh trgovcev lahko preobremenjujoča. V nadaljevanju je bilo veliko zanimanja za preučevanje prepoznavanje kriterijev, ki vplivajo na proces odločanja pri nakupovanju prek spleta. Zato smo v diplomski nalogi izvedli raziskavo in analizirali, v kolikšni meri določeni kriteriji, kot so lastnosti izdelka, ponudba izdelkov, varnost in zaupanje v spletno trgovino, ocene izdelkov, cena in popusti izdelka, ocene potrošnikov, uporabnost spletne trgovine, blagovne znamke, dostava in vračilo, vplivajo na odločitev o nakupu. Glede na dve različni starostni skupini smo ugotovili, kateri so najpomembnejši kriteriji, ki določajo odločitev o nakupu. To so: lastnosti izdelka, zaupanje v spletno trgovino, ocene izdelka, cena in popusti izdelka, dostopnost in enostavnost in ocene strank.

Logistični izzivi gostovanja ekipe NFL v Evropi: primer tekme v Münchnu : diplomsko delo

Abstract: Diplomsko delo z naslovom "Logistični izzivi gostovanja ekipe NFL v Evropi: primer tekme v Münchnu" se osredotoča na raziskavo in analizo logističnih izzivov, s katerimi se srečujejo ekipe ameriškega nogometa pri organizaciji in izvedbi mednarodnih tekem v Evropi. Glavni cilj naloge je identificirati in preučiti ključne logistične izzive, kot so prevoz ekip in opreme, prilagoditev infrastrukture, zagotavljanje ustrezne nastanitve, obvladovanje časovnih zamikov ter organizacija varnostnih ukrepov. Izbrana tekma v Münchnu bo služila kot študijski primer za podrobno analizo teh izzivov. V teoretičnem delu bomo pregledali obstoječo literaturo in teorije s področja logistike v športu, zlasti v ameriškem nogometu. Nadaljevali bomo z analizo specifičnih logističnih izzivov v ligi NFL ter primerjalno analizo med domačimi tekmami v ZDA in mednarodnimi tekmami v Evropi. Pričakovani rezultati raziskave bodo prispevali k boljšemu razumevanju logistike v športni industriji in ponudili konkretne predloge za optimizacijo logističnih procesov pri organizaciji mednarodnih športnih dogodkov. Na ta način bo diplomsko delo prispevalo k razvoju boljših praks in strategij, ki lahko olajšajo izvedbo takšnih dogodkov v prihodnosti ter izboljšajo izkušnjo za vse udeležence.

Vpliv vojne v Ukrajini na svetovni trg pšenice in zemeljskega plina: izzivi in posledice : diplomsko delo

Abstract: V diplomskem delu smo analizirali vpliv vojne v Ukrajini na svetovni trg pšenice in zemeljskega plina. Osredotočili smo se na posledice, ki jih je vojna povzročila na teh dveh ključnih trgih, in na možne rešitve za stabilizacijo oskrbe. Ugotovili smo, da je vojna močno vplivala na globalno oskrbo s pšenico, saj je Ukrajina ena največjih svetovnih izvoznic pšenice. Zaradi uničenja infrastrukture in blokade pristanišč se je ukrajinski izvoz pšenice drastično zmanjšal, kar je povzročilo dvig cen na svetovnem trgu. Podobno je vojna vplivala tudi na trg zemeljskega plina, saj je povzročila prekinitev dobav iz Rusije v Evropo, kar je sprožilo iskanje alternativnih virov energije. Predlagane rešitve vključujejo diverzifikacijo dobaviteljev, povečanje skladiščnih zmogljivosti in spodbujanje obnovljivih virov energije, kar bi prispevalo k večji stabilnosti na teh trgih v prihodnosti.
